“…EAE was induced in inbred female 7to 10-week-old Lewis rats (C.S.E.A.L., CNRS, OrlCans la Source, France) by injecting in one hind footpad 100 p1 containing 100 pg of MBP emulsified in CFA (Difco, Detroit, MI) with 2.5 x lo9 killed Bordetellu pertussis (B. pertussis) organisms (Institut Pasteur, Paris, France) Virtually every challenged rat developed severe paralysis, fecal impaction and urinary dysfunction, on days 11 to 15 with recovery from the disease on days 16 to 17; such exceptions as there were died on days 13 to 14. Rats were scored daily for typical signs of EAE following Coates [9]: no EAE = 0; flaccid tail = 1; hind leg weakness = 2; hind leg paralysis with incontinence = 3; moribund state = 4. Two preparations of MBP were used for all the experiments.…”
