Actinomyces viscosus, a normal inhabitant of the oral cavity, has previously been shown to evoke immune responses in the host. Macrophage activation by this microorganism was studied in a murine system. Peritoneal macrophages, activated in vivo by A. viscosus had a suppressive effect on lymphocyte proliferation in vitro; total suppression occurred at a PEC to lymphocyte ratio of 6:10. A. viscosus‐activated macrophages also suppressed the proliferation of normal syngeneic fibroblasts, with a 96% suppression at a PEC to fibroblast ratio of 9:1. Resident peritoneal macrophages had no suppressive effect on either target cell population. Suppression of fibroblast proliferation by A. viscosus‐activated macrophages was partially reversed by catalase (20000 U/ml) or indomethacin (10‐5M), suggesting involvement of hydrogen peroxide and cylo‐oxygenase product(s). A. viscosus‐activated macrophages produced, upon stimulation, large amounts of H2O2 (256 nmole per mg protein, in 120 min) which were comparable to those produced by C. parvum‐activated macrophages. Resident peritoneal macrophages produced only minor quantities of H2O2 under the same conditions. The suppressive effects observed suggest that A. viscosus, by activating macrophages, may modulate the host's immune response and may hamper the repair potential of chronically inflamed connective tissue by inhibition of fibroblast proliferation. Other constitutents of the plaque may share this effect.

Four hybrid cell lines secreting monoclonal antibodies against antigens of Bacteroides intermedius were generated by fusing murine NSI cells with splenocytes from a rat immunized with B. intermedius strain OMZ248. An enzymelinked immunosorbent assay was used to analyze the distribution of the recognized antigens on 39 strains from various Bacteroides species and on 5 strains from other genera. Only Bacteroides species B. intermedius, B. loescheii, B. melaninogenicus, and B. corporis were found to express at least one of the recognized antigens. Strains of the two asaccharolytic black-pigmenting Bacteroides species were negative. Among the strains capable of binding to one or more of the monoclonal antibodies, five groups with different reactivity patterns could be distinguished. Two of the monoclonal antibodies were specific for B. intermedius. The B. intermedius strains were metabolically almost identical, expressed at least three of the recognized antigens, and fell into three distinct antibody reactivity groups, suggesting a tentative separation of this species into three new serogroups. Oral and nonoral isolates of B. intermedius were, however, not distinguished by the monoclonal antibodies. One monoclonal antibody was directed against an antigen strongly expressed on all saccharolytic black-pigmenting Bacteroides strains tested so far, thus confirming the previously noted antigenic relationship between the species which had emerged from the former B. melaninogenicus subsp. intermedius and B. melaninogenicus subsp. melaninogenicus groups.

Soluble sonic extracts of several strains of Treponema denticola and Treponema vincentii were examined for their abilities to alter proliferation of both murine and human fibroblasts. We found that sonic extracts of all tested strains of T. denticola caused a dose-dependent inhibition of murine and human fibroblast proliferation when assessed by both DNA synthesis ([3H]thymidine incorporation) and direct cell counts. T. vincentii had only a minimal inhibitory effect at comparable doses. No inhibition was observed when sonic extracts were added simultaneously with [3H]thymidine, indicating that suppression was not due to the presence of excessive amounts of cold thymidine in the extract, nonspecific effects on thymidine utilization by the cells (transport and incorporation), or degradation of label. RNA ([3H]uridine incorporation) and protein ([3H]leucine incorporation) synthesis were similarly altered after exposure to the T. denticola sonic extracts. There was no effect on cell viability as measured by trypan blue exclusion. Inhibition could be reversed by extensive washing of the cells within the first few hours of exposure to sonic extracts. Preliminary characterization and purification indicated that the inhibitory factor(s) is not endotoxin since it is heat labile, and elutes in a single, well-defined peak on a Sephadex G-150 chromatography column corresponding to a molecular weight of approximately 50,000. Since oral spirochetes have been implicated in the pathogenesis of periodontal disorders, it is possible that they contribute to the disease process by inhibition of fibroblast growth and therefore may, at least in part, account for the loss of collagen seen in diseased tissue.
