Cell–Matrix Contact Prevents Recognition and Damage of Endothelial Cells in States of Heightened Immunity

Abstract: Background-Autoimmunity may exacerbate vascular disease, particularly in the form of anti-endothelial cell (EC) antibodies. The increased morbidity of cardiovascular diseases in concert with diabetes mellitus, hypertension, and other systemic illnesses may reflect the increase presence and potency of these antibodies. Matrix-embedded ECs act as powerful regulators of vascular repair accompanied by significant reduction in expected systemic and local inflammation. We compared the immune response against free … Show more

“…Endothelial cells (ECs) embedded within a 3D denatured collagen matrix secrete antiproliferative, antithrombotic, and antiinflammatory agents that promote vascular repair even when such constructs are placed far from the lumen within the vascular adventitia. Luminal inflammation, occlusive thrombosis, and intimal hyperplasia can be controlled by allo-and xenogeneic matrix-embedded (ME) ECs in a manner indistinct from native endothelium and without eliciting a significant immune response (1)(2)(3)(4)(5)(6)(7)(8). The challenge posed is whether endothelial implants recapitulate the endothelium lining as an epithelium or add to the regulation by perfusing vessels of the vasa vasorum, which are principally comprised of ECs.…”

“…porous matrices of denatured collagen retain the biosecretory capacity of quiescent endothelial cells and are immunoprotected. Perivascular matrix-embedded allogeneic endothelial cell implants in immunocompetent mice and rats did not induce endothelial-specific antibodies and showed a complete absence of the infiltration of innate and adaptive immune cells that normally cause rejection in engrafted tissues [29][30][31]. Perhaps surprisingly, the same was observed even in xenogeneic implants into immunocompetent hosts [32].…”

“…While pro-inflammatory cytokine stimulation of 2-D cultured endothelial cells resulted in this dysfunctional phenotype, matrix-embedded endothelial cells maintained a quiescent phenotype that, in fact, induces an anti-inflammatory effect in T cells and dendritic cells-two cell types that are critical to the immune response [32]. Implanted matrix-embedded endothelial cells, as compared with 2-D cultured endothelial cells, limited naïve T cell activation, differentiation, and proliferation, and decreased the differentiation of CD4 + and CD8 + T cells into effector cells, even in animal hosts with preformed antiendothelial cell immunity [29][30][31]. Furthermore, there was enhanced induction of anti-inflammatory T reg cells [35] and T h 2 cells that are responsible not just for allograft acceptance, but also for nonfibrotic tissue repair [36,37].…”

“…Sağ ve soldan yapılan ölçümlerin ortalaması KİMK olarak kaydedildi. (4)(5)(6). Üremik hastalarda da kardiyovasküler hastalığın ilk adımının endotel işlev bozukluğu olduğu ve bu patolojinin böbrek işlev kaybı ile paralel olarak arttığı kabul edilmektedir (7).…”
