Cell isolation procedures matter: a comparison of different isolation protocols of bone marrow mononuclear cells used for cell therapy in patients with acute myocardial infarction

Abstract: Cell isolation protocols have a major impact on the functional activity of bone marrow-derived progenitor cells. The assessment of cell number and viability may not entirely reflect the functional capacity of cells in vivo. Additional functional testing appears to be mandatory to assure proper cell function before embarking on clinical cell therapy trials.

“…MSC may [51], or may not [25], have greater regenerative capacity than HSC. However the lower relative numbers of non-hematopoietic stem cells [52] in the bone marrow extracted for cell-based therapy may relegate the regenerative contribution of these cells to a small role in circumstances using whole bone marrow. This characteristic also makes it difficult to use MSC in time-critical practical situations.…”

“…We carried out the BMMC injections on the same day after preparation, 8 hours after collection. Other authors have used different protocols for isolation and storage, which may have implications in the migration and retention of these cells by the myocardium 17 . This study provides evidence that the BMMC adhesion and retention via IAC injection in the infarcted myocardium is feasible and safe, confirming previous findings on the mobilization signaling and homing of stem cells by the damaged tissue, during the period of acute ischemic lesion 18 .…”

“…Seeger e colaboradores 21 demonstraram que diferenças na forma de armazenamento, na solução de coleta e armazenagem, e mesmo no gradiente de densidade podem alterar a purificação e função de células derivadas de aspirados da medula óssea. Os autores mostraram diferenças significativas na recuperação celular, especificamente de progenitores hematopoéticos (CD45…”
