Cell Elongation and Cell Death of Helicobacter pylori Is Modulated by the Disruption of cdrA (Cell Division‐Related Gene A)

Abstract: Helicobacter pylori (H. pylori), a gram-negative spiral bacterium, colonizes the human stomach and establishes a persistent infection associated with gastrointestinal inflammatory diseases including peptic gastric ulcer, duodenal ulcer, gastric cancer (31) and gastric mucosa-associated lymphoid tissue (MALT) lymphoma (2). The diversity or adaptive mutation in H. pylori genes like babA (1) generally alters the biological aspect of the microbe to response to individual conditions and eventually establishes a per… Show more

“…Sterilized water was used as a control for all experiments. The number of colony forming units (CFU) was determined as a measure of bacterial viability (Takeuchi et al, 2006). MIC was defined as the lowest AR concentration to inhibit 1 × 10 4−5 CFU mL −1 compared to controls.…”

“…Further, HPKT510 has been reported to be tolerant of ampicillin (AMC). 45,46 DNA sequencing DNA samples from the panel of H. pylori strains were subjected to polymerase chain reaction (PCR) amplification and sequencing of full-length DNA encoding the hpβCA protein. The GenBank database search identifi ed two DNA clones of hpβCA (gene symbol, icfA).…”

“…46 Briefl y, H. pylori strains were cultured overnight in brucella broth (Difco, Lawrence, KS, USA) supplemented with 10% horse serum at 37°C under microaerobic conditions; then bacterial growth was measured by determining the optical density at 600 nm (OD 600 ) with a spectrophotometer (Spectronic 20A; Shimazu, Kyoto, Japan). Following inoculation into 1.0 ml of the fresh brucella-serum medium in 2.0-ml microcentrifuge tubes at an OD 600 of 0.5 with or without sulpiride (Sigma, St. Louis, MO, USA) at fi nal concentrations of 100, 200, 300, and 500 μg/ml, they were incubated for 7 h at 37°C with shaking.…”

“…The detailed properties of the strains used were as follows: 5 strains were MNZ resistant; 2 strains were CAM resistant; and 14 strains were antibiotic susceptible. The seven isogenic mutants of H. pylori HPK5 obtained from a Japanese patient with gastric ulcer (Takeuchi et al, 1998) were as follows: cdrA-disrupted (HPKT510) (Takeuchi et al, 1998;Takeuchi et al, 2006), biotics also remain unclear. In particular, the relationship between Chinese chive (Allium tuberosum) and its functional effects on the growth of H. pylori (anti-H. pylori activity) are unknown to date.…”
