Despite the increasing number of studies on the molecular actions of quercetin and curcumin, their anticancer efficacy, safety and molecular aspects of chemopreventive action in the ovarian cancer prophylaxis or treatment and also their potential in the sensitization to cytostatics used in the clinical practice remains still not clearly understood.Based on basic studies we have summarized evidences for inhibitory activities of several often studied plantorigin bio-active compounds (mostly quercetin and curcumin) against ovarian cancer cells proliferation, their mechanisms of action as well as their strong potential to sensitization of ovarian cancer cells to the presence of several platinum-based cytostatics -cisplatin and oxaliplatin. Up to date only several dietary, clinical (cohort and case-control) studies evaluating the association of some flavonoids (mostly nonisoflavones) and its subgroup components consumption and ovarian cancer risk were already performed. According to the researchers, there has been no association between ovarian cancer risk and total nonisoflavone flavonoids intake. There is a still an insufficient amount of data designed to explain the effect of quercetin or curcumin (alone or together) on ovarian cancer development and/or its chemotherapy. Obtained results provide limited support for an association between nonisoflavone flavonoids intake and ovarian cancer risk, therefore there is a need for further and more accurate studies to be confirmed. We are of the opinion that this paper will contribute to a better understanding of the molecular basis for positive interactions between concomitant usage of quercetin or curcumin with above-mentioned cytostatics and other bio-active agents. This work may also contribute to an increase in the number of preclinical studies or other clinical, dietary trials using these or other phenolic / alkaloid, plant-origin constituents in order to investigate efficiency and safety of pharmacotherapy of ovarian cancer patients.

Asian Pacific J Cancer Prev, 13, 289-294 IntroductionOral submucous fibrosis (OSF) is a chronic inflammatory disease, and has been defined by WHO as one of the precancerous conditions (International Agency for Research on Cancer, 2005). The main clinical symptoms include the stiffness of oral mucosa, restriction of mouth opening, atrophy of tongue papillae, blisters, and intolerance to hot and spicy food. Malignant transformation rate in OSF is high, reaching 7-13% (Tilakaratne et al., 2006). The disease exhibits characteristic histological features of excessive collagen deposition in the lamina propria, following the epithelium becomes atrophic (Singh et al., 2010).OSF is closely associated with betel quid (BQ) chewing, a habit common in South and Southeast Asia, some parts of Africa, and among immigrants from these regions (Boucher and Mannan, 2002). An imbalance in collagen synthesis and degradation in oral mucosa is generally believed to be the main cause of OSF;The disease is considered a type of collagen metabolism disorder (Rajalalitha and Vali, 2005).Myofibroblasts are typically activated fibroblasts, although they can also be derived from other cell types, including epithelial and endothelial cells via epithelial/ endothelial mesenchymal transition (EMT/EndMT) process, as well as from the circulating fibroblast-like cells AbstractBackground: Myofibroblasts play an important role in the development of oral submucous fibrosis (OSF). In the current study, we investigate the effect of curcumin on growth and apoptosis of myofibroblasts derived from human oral mucosa. Methods: Myofibroblasts were generated by incubating fibroblasts, obtained from human oral mucosa, with transforming growth factor-β1 (TGF-β1). MTT, PI staining, and FACS assays were used to investigate curcumin's effect on proliferation and cell cycle of fibroblasts and myofibroblasts. Annexin V/PI binding and FACS assays were used to examine apoptosis of myofibroblasts, Western blotting to determine the levels of Bcl-2 and Bax, and enzyme-linked immunosorbant assay was employed to examine the levels of collagen type I and III in the supernatants of myofibroblasts. Results: Curcumin inhibits proliferation of fibroblasts and myofibroblasts; it also disturbs the cell cycle, induces apoptosis and decreases the generation of collagen type I and III in myofibroblasts, which are more sensitive to its effects than fibroblasts. Curcumin induces apoptosis in myofibroblasts by down-regulating the Bcl-2/ Bax ratio. Conclusion: Our results demonstrate the antifibrotic effect of curcumin in vitro. It may therefore be a candidate for the treatment of OSF.

Objective. Curcuma longa (C. longa) was used in some countries such as China and India for various medicinal purposes. Curcumin, the active component of C. longa, is commonly used as a coloring agent in foods, drugs, and cosmetics. C. longa and curcumin have been known to act as antioxidant, anti-infl ammatory, anti-mutagen, and anti-carcinogenic agents. Th e attempt of the present review was to give an eff ort on a detailed literature survey concentrated on the protective eff ects of C. longa and curcumin on the reproductive organs activity.Methods. Th e databases such as, PubMed, Web of Science, Google Scholar, Scopus, and IranMedex, were considered. Th e search terms were "testis" or "ovary" and "Curcuma longa", "curcumin", "antioxidant eff ect", "anti-infl ammatory eff ect" and "anti-cancer eff ect".Results. C. longa and curcumin inhibited the production of the tumor necrosis factor-α (TNF-α) and prostaglandin E2 (PGE2) and increased the caspases (3, 8 and 9) activities in HL-60 prostate cancer. Furthermore, C. longa and curcumin suppressed the vascular endothelial growth factor (VEGF), phosphorylated signal transducers and activators of the transcription 3 (STAT) and matrix metalloproteinase-9 (MMP-9) in ovarian cancer cell line.Conclusion. C. longa and curcumin might decrease the risk of cancer and other malignant diseases in the reproductive system. C. longa and curcumin have a protective eff ect on the reproductive organs activity such as, anti-infl ammatory, anti-apoptotic, and antioxidant eff ects in normal cells but showed pro-apoptotic eff ects in the malignant cells. Th erefore, diff erent eff ects of C. longa and curcumin are dependent on the doses and the type of cells used in various models studied.
