Celastrol and Melatonin Modify SIRT1, SIRT6 and SIRT7 Gene Expression and Improve the Response of Human Granulosa-Lutein Cells to Oxidative Stress

Abstract: An excess of oxidative stress (OS) may affect several physiological processes fundamental to reproduction. SIRT1, SIRT6 and SIRT7 are involved in protection stress systems caused by OS, and they can be activated by antioxidants such as celastrol or melatonin. In this study, we evaluate SIRT1, SIRT6 and SIRT7 gene expression in cultured human granulosa-lutein (hGL) cells in response to OS inductors (glucose or peroxynitrite) and/or antioxidants. Our results show that celastrol and melatonin improve cell surviva… Show more

“…The sirtuin family consists of seven proteins (encoded by the genes SIRT1, SIRT2, SIRT3, SIRT4, SIRT5, SIRT6, and SIRT7), which deacetylate histones in a nicotinamide adenine dinucleotide (NAD + )-dependent manner and which are involved in the repair of damaged DNA, too [ 7 ]. In addition, the expression of sirtuins can be increased by supplementing a stilbene derivative, i.e., resveratrol, melatonin, astaxanthin, celastrol, or mogroside [ 8 , 9 , 10 , 11 ]. The activators presented have been touted for improving oocyte quality or reversing the effects of oxidative stress, and some, such as melatonin, have been proposed as candidates for the composition of media in in vitro cultures.…”

“…Specifically, natural compounds 1 , 3 , 4 , , 5 , 8 , , 9 , and drug 11 , demonstrate the capability to also activate SIRT1. Compound 7 exhibits the direct induction of the SIRT7 gene expression, whereas compound 13 elicits the expression of SIRT1, SIRT6, and SIRT7 . However, all these compounds ( 1 – 15 ) could be considered potential lead compounds to be optimized through a med-chem approach.…”

“…SIRTs regulate metabolism, energy expenditure, and the aging process which, in turn, are regulated by diet and environmental stress [25][26][27][28]. SIRTs are found in the GCs of various mammalian species including humans at different stages of the reproductive cycle [21,[29][30][31][32][33][34][35].…”
