cDNA cloning, expression and characterization of an allergenic L3 ribosomal protein of Aspergillus fumigatus

Abstract: SUMMARYAspergillus fumigatus ( Afu ) is an important fungal pathogen causing allergic and invasive respiratory disorders. A plethora of multi-functional allergens/antigens secreted by Afu have been implicated in pathogenesis. The present study was undertaken to identify and characterize novel Afu allergen/antigen by cDNA library approach. cDNA library of Afu was immunoscreened with pooled sera of allergic bronchopulmonary aspergillosis (ABPA) patients. The cDNA clone, TS1, reacting significantly with specific … Show more

“…The ability of P1 to stimulate histamine release from ABPA patients and cytokines of Th2 type, although at a lower magnitude than SDA, suggests that it could be further explored for intradermal skin testing. There have been numerous reports of diagnostically relevant native and recombinant antigens for serodiagnosis of aspergillosis (12,14,15,16,19,25,28,30). The reported diagnostic sensitivity and specificity vary greatly among the different reports, even for similar antigen preparations.…”

“…High throughput screening of a human lung cDNA library displayed on phage surface [31, 32] revealed more than 150 partial and complete cDNA sequences potentially encoding IgE-binding proteins. Although not yet analyzed in detail, some of the cDNA sequences, like those encoding α-tubulin [33] or ribosomal L 3 protein [34], show a high degree of sequence identity and cross-reactivity to environmental allergens [Zeller et al, unpublished results]. In these cases, it makes sense to assume that IgE autoreactivity can be traced back to common structural features shared between environmental and self antigens (see below).…”

“…Molecular biology techniques were previously used for the identification of Ags and allergens from various fungal species such as Aspergillus fumigatus and Fusarium culmorum (16,17). In analogy to these studies, we now searched a newly developed M. mycetomatis cDNA expression library for immunodominant Ags.…”
