CDK5RAP1 deficiency induces cell cycle arrest and apoptosis in human breast cancer cell line by the ROS/JNK signaling pathway

Abstract: Abstract. Cyclin-dependent kinase 5 regulatory subunit associated protein 1 (CDK5RAP1) is an enzyme which post-synthetically converts the RNA modification N6-isopentenyladenosine (i 6 A) into 2-methylthio-N6-isopentenyladenosine (ms 2 i 6 A). However, the interaction between CDK5RAP1 deficiency and cell apoptosis has not been studied. Breast cancer has long been a leading cause of mortality in the world. Therefore, in the present study, CDK5RAP1 deficiency in a human breast cancer cell line was investigated. C… Show more

“…CDK5RAP1 also acts as an inhibitor of CDK5 to maintain healthy neuronal development and neurogenesis (37). CDK5RAP1 deficiency suppresses tumor growth and induces cell cycle arrest and apoptosis in human breast cancer (38). Although 2MeS-CKs were detected in our study (2MeSiPR, 2MeSiP, and 2MeSZR), their tissue specificity and function in mammalian physiology, beyond tRNA modification, have yet to be investigated.…”

“…In the past, surgery and radiotherapy has been the most common treatment method ( 3 ). Although there is now a new era of molecular targeted therapy, these traditional therapies often bring numerous unwanted side effects and problems, and do not always improve the symptoms ( 4 ). Due to the limited efficacy traditional therapeutics, it is important to identify novel treatment strategies with reduced side effects.…”

“…Previous studies showed that HS6ST2 was involved in ovarian cancer (Cole et al 2014) and pancreatic cancer (Song et al 2011). CDK5RAP1 was associated with tumor growth in breast cancer (Wang et al 2015). Though tubal EM is not a malignant disease, some biological behaviors of it are similar to cancer, including invasion, adhesion, angiogenesis and proliferation.…”
