CDK12/13 inhibition induces immunogenic cell death and enhances anti-PD-1 anticancer activity in breast cancer

Abstract: Background: The T cell response against different tumors is improved by immunogenic cell death (ICD), indicating a role for ICD in augmenting antitumor immunity elicited by the anti-checkpoint antibody antiprogrammed death 1 (anti-PD-1). Methods: The effect of SR-4835 on breast cancer was analyzed by cell proliferation and ow cytometry. Calreticulin translocation and HMGB1 and ATP release were detected by ow cytometry, ELISA and luminescence assay, respectively. Immunogenicity in vitro was analyzed by co-cultu… Show more

“…Delivery of ICD inducers, such as SR-4835 (a CDK12/13-specific inhibitor), RIG-I agonists and chemotherapeutics, as well as photothermal agents, with synthetic nanoparticles, has been confirmed to have therapeutic potential in several preclinical models of breast cancer [45][46][47]. Although Texs play a major role in tumor immune evasion and growth, they can be manipulated and have tumor-homing properties, as well as low in vivo toxicity and immunogenicity [48].…”

“…The notion of immunogenic cell death was described as the unique type of regulated cell death, able to trigger full antigenspecific adaptive immunological responses by emitting danger signals or DAMPs (1,3,14). The combination of immunogenic therapeutic and novel immunotherapeutic regimens holds great promise for treating malignancies (15)(16)(17)(18). Therefore, it could be advantageous to identify ICD-related biomarkers that help distinguish HNSCC patients premised on the benefits they derive from immunotherapy.…”

“…Several recent clinical reports now corroborate this hypothesis as the sequential combination of ICD induction by chemo- or radiotherapy with PD-1 checkpoint blockade achieved excellent therapeutic efficacy in several neoplastic indications including breast and small cell lung cancer. 17–23 …”
