CD80 Expression on Tumor Cells Alters Tumor Microenvironment and Efficacy of Cancer Immunotherapy by CTLA-4 Blockade

Abstract: Cluster of differentiation (CD) 80 is mainly expressed in immune cells but can also be found in several types of cancer cells. This molecule may either activate or inhibit immune reactions. Here, we determined the immunosuppressive role of CD80 in the tumor microenvironment by CRISPR/Cas9-mediated deactivation of the corresponding gene in the mouse oncogenic TC-1 cell line. The tumor cells with deactivated CD80 (TC-1/dCD80-1) were more immunogenic than parental cells and induced tumors that gained sensitivity … Show more

“…Kallikrein-related Peptidase 9 (KLK9) , reported as a favorable prognostic marker in ovarian cancer [ 55 ], was highly upregulated by the combination of drugs, as was Orosomucoid 2 ( ORM2 ), a tumor suppressor in liver cancer [ 56 ]. Nonetheless, the other upregulated genes have been associated to pro-tumorigenic roles and poor prognosis: SIGLEC-11 [ 57 ], CAMP [ 58 ], CD80 [ 59 ], and CLIC6 [ 60 ].…”

“…We suspect that immune system and tumor microenvironment (TME) were extremely complex, involving multiple cells and substances. ICIs regulated not only cytotoxic T cells but also regulatory T cells, macrophages, helper T cells, natural killer cells, dendritic cells, and bone marrow-derived suppressor cells [ 31 ]. A single factor could not perfectly and systematically reflect the immunity, inflammation, and nutritional status of the person.…”

“…CD80 is an essential membrane antigen for the activation of T lymphocytes. CD80 monoclonal antibody inhibits the adjuvant stimulation of CD80, and prevents the differentiation of B lymphocytes into plasma cells, which plays a prominent role in the treatment of tumors (Vackova et al, 2021). CD27 and CD40 belong to the tumor necrosis factor receptor (TNFR) family.…”
