“…Recently, we quantified colocalization of CD74 and CD44 in breast cancer cells through noninvasive and validated bioimaging procedure [28] and also determined several novel biomarkers involved in the pathogenesis of breast cancer [29]. Not only have these, but we also showed that treatment of human breast cancer cells with interferon-γ up-regulates the expression of CD74 along with MIF and CD44 [2], [30]. In continuation of these studies, the present study was hypothesised to find out the potential roles of CD74 in the proliferative and apoptotic activity in breast cancer.…”

“…We validated the specificity of UMAb231 for IHC using the human breast cancer cell line MDA-MB-231. This cell line has a robust endogenous expression of CD74 protein, which can be easily upregulated with IFNγ or suppressed with siRNA [ 35 , 39 , 56 ]. Western blot analysis of whole cells extracts with endogenous expression (control lanes; Figure 1 A), siRNA-mediated depletion- (+CD74 siRNA lanes; Figure 1 A) or IFNγ-mediated up-regulation (+IFNγ lanes; Figure 1 A) of CD74, showed that the LN-2 antibody recognized a major band at 35 kDa, congruent with the expected molecular weight for CD74.…”

“…Bones were also collected from the experimental rat's groups and the acquired bone marrow was harvested to isolate and culture MSCs for osteoblastic differentiation. MSCs (passage 3) were tested for viability by trypan blue stain and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT), (Sigma-Aldrich, USA) assays before culturing in osteogenic media as described previously (Alabdulmonem et al, 2018;Rasheed et al, 2010). The cell suspension was done at an appropriate concentration of 1 × 10 5 /ml in α-MEM as described previously (Quan et al, 2016).…”
