CD44 Interaction with c-Src Kinase Promotes Cortactin-mediated Cytoskeleton Function and Hyaluronic Acid-dependent Ovarian Tumor Cell Migration

Abstract: In this study we have demonstrated that both CD44 (the hyaluronan (HA) receptor) and c-Src kinase are expressed in human ovarian tumor cells (SK-OV-3.ipl cell line), and that these two proteins are physically associated as a complex in vivo. Using a recombinant cytoplasmic domain of CD44 and an in vitro binding assay, we have detected a specific interaction between CD44 and c-Src kinase. Furthermore, the binding of HA to SK-OV-3.ipl cells promotes c-Src kinase recruitment to CD44 and stimulates c-Src kinase ac… Show more

“…Assessment of anoikis and treatment of inhibitor Cells were prevented from adhering to the plastic dishes by culturing them in dishes coated with PolyHEMA (Sigma, St Louis, MO, USA) as described previously (Bourguignon et al, 2001). Briefly, culture of transient or stable-transfected cells was tripsinized and plated onto 6-well polyHEMA plates (polyHEMA plates were made by applying 1.5 ml of 10 mg/ml solution of polyhydroxyethylmethacrylate in ethanol onto the plate, drying in tissue culture hood).…”

“…Furthermore, crosslinking of CD44 results in higher expression levels of lymphocyte function-associated antigen-1 (LFA-1) and VLA-4 which are responsible for enhanced transendothelial migration of tumor cells [38]. The cytoplasmic tail cooperates with intracellular molecules as members of the Src tyrosine kinase family including c-Src, Lyn, Fyn, and Lck [42][43][44][45], Moesin-Ezrin-Radixin-Like Protein Merlin [46], T-cell lymphoma invasion and metastasis-inducing protein 1 and 2 (Tiam1/2) [47], intracellular OPN [48], as well as members of the Smad protein family [14,49,50].…”

“…Besides direct interactions, CD44 influences filament organization by co-activation of several tyrosine kinases resulting in downstream phosphorylation of cytoskeleton proteins [43] and F-actin rearrangement [44].…”

“…[16][17][18] In particular, tyrosine phosphorylation of E-cadherin and b-catenin induces their disassembly from adhesion junctions. We therefore investigated whether the observed increase in cell-cell and cell-substratum adhesions was dependent on an inhibition of c-Src kinase after NAC treatment.…”
