CD40 expression by human monocytes: regulation by cytokines and activation of monocytes by the ligand for CD40.

Abstract: SummaryCD40 is a member of the tumor necrosis factor (TNF) receptor family of cell surface proteins and was originally described as a B cell restricted antigen. Treatment of primary human monocytes with granulocyte/macrophage colony-stimulating factor (GM-CSF), interleukin 3 (IL-3), or interferon 3' (IFN-3') resulted in the induction of CD40 mRNA and enhancement of cell surface protein expression. CD40 was found to mediate monocyte adhesion to cells expressing recombinant CD40 ligand. CD40 ligand-transfected c… Show more

“…26 In addition, CD40 ligation on macrophages or monocytes enhances their TNF -production, and induces tumoricidal activity. 27 We have previously demonstrated that the 3LLSA -CD40L used in this study could activate murine alveolar macrophages and enhance their tumoricidal activity in vitro. 11 It is also known that CD40 ligation inhibits spontaneous or tumor-induced apoptosis of dendritic cells.…”

“…One is that macrophages or dendritic cells at the tumor site are activated through CD40 ± CD40L interaction, and they release cytokines and mediators such as TNF -or nitric oxide to kill tumor cells. 27 They also release IL-12 to recruit NK cells those are important for eliciting local antitumor response. 32 Our imunohistochemical study showing the enhanced infiltration of NK cells into 3LLSA -CD40L tumor may support this scenario.…”

“…Previous studies have shown that TNF-, IFN- and IFN- upregulate CD40 expression in various cell types including monocytes [24,25]. NF-B and IRF-1 have been implicated in the regulation of CD40 expression [26][27][28].…”
