CD4+CD25+T Cells Prevent Arthritis Associated withBorreliaVaccination and Infection

Abstract: CD4؉ CD25 ؉ T cells are a population of regulatory T cells associated with control of arthritis in antiinterleukin-17 antibody-treated Borrelia-vaccinated and challenged gamma interferon-deficient mice. Here, we present direct evidence that adoptive transfer of enriched CD4 ؉ CD25 ؉ T cells from these mice can prevent the development of arthritis in Borrelia-vaccinated and challenged mice. These findings establish a major role for CD4؉ CD25 ؉ T cells in the prevention of arthritis in Borrelia-vaccinated and ch… Show more

“…The data we present here are reminiscent of our findings linking IL-17 and putative Treg cells in a model of Borrelia-induced arthritis (13,42,43). We showed that blocking IL-17 in IFN-␥-deficient, Borrelia-vaccinated and -infected mice prevented pathology and increased the production of CD25-expressing CD4 ϩ cells (13).…”

“…In fact, we have recently used US to evaluate knee involvement in a series of 30 consecutive Italian Behçet's syndrome patients, and the results demonstrated a high prevalence of signs of synovitis even among patients without symptoms (8). These evaluations confirmed the high sensitivity of US for the detection of joint alterations in rheumatic diseases and its role in detecting subclinical inflammation.…”

“…Administration of anti-CD25 antibody in these anti-IL-17 antibody-treated, Borreliavaccinated and -infected mice reduces the number of CD4ϩ CD25ϩ T cells in the local lymph nodes, and more importantly, results in the development of severe, destructive arthritis (7). In addition, the transfer of enriched CD4ϩCD25ϩ T cells obtained from anti-IL-17 antibody-treated, Borrelia-vaccinated and -infected mice into untreated Borrelia-vaccinated and -infected mice prevents the development of pathologic changes (8), suggesting that these cells have immunoregulatory properties. These studies need confirmation in humans.…”
