Psoriasis is a relatively common disorder in children and can be triggered by an upper respiratory tract infection. The aim of this study was to compare the clinical features of psoriasis in children and adult. In addition, we evaluated the relationship between anti-streptolysin O (ASO) titers and the clinical features of psoriasis. A total of 30 childhood psoriasis patients and 30 adult psoriasis patients were evaluated. Childhood psoriasis had a facial predominance when compared with the adult psoriasis. The childhood psoriasis patients with high ASO titers had guttate psoriasis more frequently than patients with normal ASO titers. In children with plaque-type psoriasis, psoriasis area and severity index score was increased in the high ASO titer group than normal ASO titer group. In conclusion, if the children with psoriasis show increased ASO titer, the physician should pay attention to the worsening of the psoriasis. Furthermore, early treatment of streptococcal infections might be beneWcial in childhood psoriasis.

Development of Cytomegalovirus and Adenovirus-Specific Memory CD4 T-Cell Functions From Birth to Adulthood

Age-related changes in memory CD4ϩ T cells (CD4) are poorly known. To address this issue, CD4 proliferative and cytokine responses to an anti-CD3 monoclonal (CD3), to cytomegalovirus (CMV), and to adenovirus (AdV) were assessed in 57 children (age, 0.07-17.16 y) and 17 adults. Results showed i) accumulation of memory CD4 with aging, with 2-3 times more centralmemory T cell (TCM; CD45RA Ϫ /CD62L ϩ ) than effector-memory T cell (TEM; CD45RA Ϫ /62L Ϫ ) CD4 at any age. ii) In children older than 2 y, CMV-specific CD4-secreting IFN␥ alone predominated over CD4-secreting IL2 ϩ IFN␥ and a continuous increase, with aging, in IFN␥ responses to the virus was observed. In contrast, in AdV infection, CD4-secreting IL2 ϩ IFN␥ predominated and IFN␥ responses to the virus reached adult levels from 3 y of age. iii) In children aged 0 -2 y, lower total IFN␥ responses to CMV (p Ͻ 0.02), AdV (p ϭ 0.05), and CD3 (p Ͻ 0.01) and lower IFN␥ ϩ IL2-responses (p ϭ 0.1, p Ͻ 0.02, p Ͻ 0.05, respectively) contrasted with no decrease in CD4-secreting IFN␥ alone. Defective proliferative responses to AdV (p ϭ 0.03) were also observed. In conclusion, the development of memory CD4 differed in acute AdV and persistent CMV infections. Young age seemed to depress mostly polyfunctional (IL2 ϩ IFN␥ secreting) CD4 in both infections. (Pediatr Res 69: 106-111, 2011)

Our objective was to study the effects of type I insulin-like growth factor receptor (IGF-IR) on human cord blood lymphocyte (CBL) functions. First, we used RT-PCR to determine the expression of IGF-IR at the mRNA level in CBL. We then inhibited the expression of IGF-IR in CBL by the antisense oligonucleotide for the IGF-IR gene. We measured the changes in interleukin (IL)-2, -4 and interferon-γ (IFNγ) at mRNA levels by RT-PCR, immunoglobulin M (IgM) production by CBL with an ELISA and lymphocyte proliferation by a 3 H-thymidine uptake technique. Our results showed that IGF-IR mRNA was detected in both non-activated and activated CBL, but the expression levels in the activated CBL were higher than those in the non-activated CBL. After being exposed to the antisense oligonucleotide, a 50% reduction in the amount of IGF-IR mRNA occurred. Accordingly, the proliferation of CBL to mitogen was significantly reduced about 50%, and the production of IgM from CBL was also markedly decreased. In the phytohemagglutinin-stimulated CBL culture system, when the IGF-IR antisense oligonucleotide existed, the mRNA levels of IFNγ and IL-2 decreased 30-50% and IL-4 decreased 20-30%. We concluded that IGF-IR is most likely involved in the process of CBL proliferation and production of immunoglobulin and cytokines. It might therefore play an important role in the modulation of the immune functions.
