CD21 antigen in T‐lineage neoplastic lymphoid cells: Characteristic expression at thymic stage

Abstract: The expression of CD21 antigen, a receptor for the C3d fragment of complement and Epstein-Barr virus (EBV), was investigated in a total of 85 cases of neoplastic lymphoid cells including 39 cases of T-lineage acute lymphoblastic leukemia (ALL)/lymphoblastic lymphoma (LBL), although CD21 antigen is usually regarded as a pan-B antigen. The CD21 antigen was expressed by one of the eight cases of neoplastic lymphoid cells expressing the CD7 antigen as a sole pan-T antigen, by three of the 20 cases of pro- or early… Show more

“…However, earlier studies showed that T‐acute lymphoblastic leukemia (ALL) cell line (MOLT4), null cells, and undifferentiated T‐cells also expressed functional C3d‐receptors and are able to bind to EBV 42,43 . In addition, Tatsumi et al 44 . detected expression of CD21, which corresponds to the C3d component of EBV receptor, in 80% (8/10) of thymic stage T‐cell ALL/lymphoblastic lymphoma (LBL), but not in pro‐ or early thymic T‐ALL/LBL.…”

Therefore, EBV may infect these cells by binding to the C3d component of the EBV receptor.…”

“…17 Usually TdT is dominantly expressed in most immature hematopoetic cell stage, eg in pro T-cell / pro B-cell stage of maturation, 18,19 but some previous studies reported that in thymic stage (CD3-CD4-CD8, CD3-CD4+CD8+, CD3+CD4±CD8± lymphoblast) TdT was still almost always expressed. 16,17,20 Negative TdTs are more easily found in CD7+CD5-CD2-lymphoblast. 16,17,20 Our negative TdT cases might be from positive myeloid marker expressingblasts and the only one positive TdT case was negative myeloid marker expressing-blast.…”

More samples are needed to get the similar conclusion as previously reported that TdT transcripts in myeloid marker + ALL is reduced than in myeloid marker -ALL.…”

“…As we have shown, the gene expression of RAG-1 (recombination activating gene-1) is markedly activated at this stage, leading to the most diverse clonal expansion by rearrangement of the TCR-P chain gene [41]. CD21 antigen also begins to be expressed mostly at this stage [42]. A multitude of cellular mechanisms must be pre-pared.…”
