CD1d deficiency exacerbates inflammatory dermatitis in MRL‐lpr/lpr mice

Abstract: Mechanisms responsible for the development of autoimmune skin disease in humans and animal models with lupus remain poorly understood. In this study, we have investigated the role of CD1d, an antigen-presenting molecule known to activate natural killer T cells, in the development of inflammatory dermatitis in lupus-susceptible MRL-lpr/lpr mice. In particular, we have established MRL-lpr/lpr mice carrying a germ-line deletion of the CD1d genes. We demonstrate that CD1d-deficient MRL-lpr/lpr mice, as compared wi… Show more

“…The role of NKT cells in lupus is controversial (45,(53)(54)(55)(56)(57)(58). A reduced level of NKT cells is observed at the spontaneous onset of lupus in the MRL/lpr mouse strain and elimination of NK1.1 cells or the CD1d gene exacerbated disease progression.…”

“…CD1d deficiency in SLE model mice aggravates the disease symptoms [7,8]; thus, CD1d could be one of the pathogenic factors of SLE. Although the decreased CD1d cell surface expression on B cells was reported in only SLE patients [27], the expression of CD1d in B cells in SLE-like mice is unclear.…”

“…Several studies have suggested that CD1d may play an immunoregulatory role in the development of lupus, and CD1d deficiency exacerbates lupus nephritis in the pristineinduced model [7] and inflammatory dermatitis in MRL-lpr/lpr mice [8]. Importantly, endogenous DNA stimulates inflammatory responses through TLR9 and exacerbates lupus disease pathology [9,10].…”

“…In support of this idea, several studies have demonstrated that iNKT cells are important to generate the protective immunity against specific parasitic or bacterial infections (Chackerian et al, 2002;Gonzalez-Aseguinolaza et al, 2000). In addition, iNKT cells have an anti-cancer activity (Kawano et al, 1998) and are used as the therapeutics to treat several autoimmune diseases Yang et al, 2004). Therefore, the regulation of iNKT cell activity is considered to be a key to the development of immunotherapy against human diseases.…”
