CD133-expressing thyroid cancer cells are undifferentiated, radioresistant and survive radioiodide therapy

Abstract: Purpose131I therapy is regularly used following surgery as a part of thyroid cancer management. Despite an overall relatively good prognosis, recurrent or metastatic thyroid cancer is not rare. CD133-expressing cells have been shown to mark thyroid cancer stem cells that possess the characteristics of stem cells and have the ability to initiate tumours. However, no studies have addressed the influence of CD133-expressing cells on radioiodide therapy of the thyroid cancer. The aim of this study was to investiga… Show more

“…In the present study, such spheres maintained an undifferentiated state (UPTC), as confirmed by morphological analysis (Fig. 1A) and the expression of stem cell markers such as CD133 (data not shown), as found in previous studies (17,22). Morphological analysis revealed that these cell cultures were exclusively formed by cellular clusters resembling so called 'tumor spheres'.…”

“…Hong et al previously reported that SUV max greater than 3.6 in distant metastases from DTC was significantly predictive of reduced disease-specific survival in multivariate analysis [19]. All of the patients with SUV max greater than 3.8 for LM in our study showed an increased TG level after and high resistance to 131 I therapy [20]. Therefore,…”

“…Conventional therapies target mature cancer cells and thus, thyroid CSCs, which are relatively quiescent and intrinsically resistant, are not eradicated (74,75). CSCs efficiently repair DNA damage following exposure to cytotoxic injury, and are therefore capable of reconstituting the original tumor (74).…”
