Ccl2 and Ccl3 Mediate Neutrophil Recruitment via Induction of Protein Synthesis and Generation of Lipid Mediators

Abstract: Objective-Although the chemokines monocyte chemoattractant protein-1 (Ccl2/JE/MCP-1) and macrophage inflammatory protein-1␣ (Ccl3/MIP-1␣) have recently been implicated in neutrophil migration, the underlying mechanisms remain largely unclear. Methods and Results-Stimulation of the mouse cremaster muscle with Ccl2/JE/MCP-1 or Ccl3/MIP-1␣ induced a significant increase in numbers of firmly adherent and transmigrated leukocytes (Ͼ70% neutrophils) as observed by in vivo microscopy. This increase was significantly … Show more

“…In contrast, RBCs themselves appear to possess the ability to scavenge inflammatory mediators, 58 a property that may be lost during storage. Consistent with this, aged RBCs display reduced capacity to scavenge CCL2, 61 a chemokine capable of recruiting neutrophils in vivo, 62 probably because of altered ligand recognition and reduced expression of Duffy antigen 61 (Figure 4). Taken together, these results importantly illustrate that several transfusion-associated factors can significantly modulate neutrophil function, with obvious implications in the development of TRALI (Figs.…”

“…MCP-1 recruits monocytes and neutrophils through the CCR2 receptor which is constitutively expressed at high levels on monocytes and is up-regulated on neutrophils in response to injury. 34,35 IL-1␤ activates monocytes and neutrophils secrete TNF-␣, IL-1␤, and MCP-1.…”

“…The higher concentration of such cytokines might additionally explain the higher content of CD11b + Gr1 + neutrophils in tumors from Ifnb1 -/-mice and their augmented growth. CCL2 is additionally known to be a potent chemoattractant for myeloid cells, such as monocytes and neutrophils (39,(50)(51)(52)(53)(54). It is believed to be produced by various cells, including vascular smooth muscle cells (55,56).…”
