Caveolae Mediate Growth Factor-induced Disassembly of Adherens Junctions to Support Tumor Cell Dissociation

Abstract: Remodeling of cell-cell contacts through the internalization of adherens junction proteins is an important event during both normal development and the process of tumor cell metastasis. Here we show that the integrity of tumor cell-cell contacts is disrupted after epidermal growth factor (EGF) stimulation through caveolae-mediated endocytosis of the adherens junction protein E-cadherin. Caveolin-1 and E-cadherin closely associated at cell borders and in internalized structures upon stimulation with EGF. Furthe… Show more

“…86 In cancer cells, EGF can induce caveolin-1-mediated degradation of Ecadherin and tumor cell dissociation. 87,88 Reggie/flotillins are microdomain scaffolding proteins (lipid raft components) that play an important role in Wnt and Shh signaling 89 and are involved in regulating membrane trafficking and turnover. 90,91 In association with prion protein (PrP), reggies/ flotillins control activation of Src kinases and the presence of receptor tyrosine kinases such as EGFR at the membrane.…”
