Causal Link between n-3 Polyunsaturated Fatty Acid Deficiency and Motivation Deficits

Abstract: Highlights d Developmental deficiency in n-3 PUFA lipid species leads to motivational deficits d n-3 PUFA deficiency increases D2-MSN lateral inhibition in the nucleus accumbens d Rescue of n-3 PUFA levels in D2 neurons normalizes lateral inhibition of D2-MSNs d Rescue of PUFA levels in D2 neurons enhances motivation under n-3 PUFA deficiency

“…Knowing the major importance of the brain in regulating food intake, this study was the first step which enable to understand the alteration of growth performance of trout fed with 100% plant based-diet devoid of u-3 LC-PUFAs. In this way, recent study demonstrated a causal link between a behavioral deficit and u-3 LC-PUFAs decrease in a discrete brain cell population suggesting that lower u-3 LC-PUFAs biostatus could participate in the etiology of reward-related symptoms and then could influence food-reward [50]. This study demonstrated that all juveniles reached the same final body weight, with no differences in growth parameters (Table 3).…”

“…During thawing, samples were treated with protease inhibitors (Complet ultra tablet, Roche Diagnostics, Meylan, France) and buffered with NaOH 1 mol/L to adjust the acidic pH of the extraction buffer. Then, equal quantities of proteins were electrophoresed on 12% sodium polyacrylamide-dodecyl sulfate gel with a 4% stacking gel and transferred to nitrocellulose membranes (Amersham 300 × 4 mm, UGAP, Champssur-Marne, France) as previously described [53]. Membranes were saturated 1 h in 5% milk (Regilait, UGAP, Champs-sur-Marne, France) or 5% BSA (P06-139,1100, Dutscher, Montsaunès, France) in Tris-Buffered Saline (TBS) and Tween 0.1%, then incubated with different primary antibodies overnight at 4 • C: anti-DRD1 (D2944; Sigma, Molsheim, France), anti-DRD2, anti-DAT (AB5084P, AB2231 respectively; Millipore, Molsheim, France), anti-SERT, anti-5-HTR1a (ab172884, ab85615 respectively; Abcam, Cambridge, UK) and anti-GAPDH (glyceraldehyde 3-phosphate dehydrogenase) as maintenance protein (5174S, rabbit; Cell Signaling, Leiden, The Netherlands).…”

“…The antagonistic effects of D1-and D2-MSNs of the NAc in the modulation of motivational processes in the context of food-seeking behavior have been recently challenged. Here, we found that chemogenetic activation or inhibition of D2-MSNs did not impact licking patterns during consumption of a palatable solution (SF2), a proxy for hedonic reactivity (Johnson, 2018a;Ducrocq et al, 2019Ducrocq et al, , 2020, but decreased and increased performance in a progressive ratio task, respectively (Figure 2A-D; SF2), without alteration in motoric abilities (Figure 2E, SF2), supporting an "amotivational" effect of D2-MSNs of the NAc core.…”

“…Bursts were defined as sequences of high frequency licks, in which the interlick interval was less than 250ms. Relation between number of licks per burst and hedonic reactivity have been demonstrated in (Ducrocq et al, 2020;Johnson, 2018b;Ward et al, 2012).…”
