Caught red handed: modeling and confirmation of the myeloperoxidase ceruloplasmin alpha-thrombin complex

Abstract: The work is devoted to the study of the structural characteristics of the myeloperoxidase–ceruloplasmin–thrombin complex using small-angle neutron scattering methods in combination with computer modeling, as well as surface plasmon resonance and solid-phase enzyme assay. We have previously shown that the functioning of active myeloperoxidase during inflammation, despite the presence in the blood of an excess of ceruloplasmin which inhibits its activity, is possible due to the partial proteolysis of ceruloplasm… Show more

“…It is to be noted that binding with MPO did not result in the inhibition of enzyme activity for all forms of ceruloplasmin. Breaking of one bond in the polypeptide chain could be enough for the loss of inhibitory potential of ceruloplasmin towards MPO even though the efficacy of its binding with MPO was not influenced [ 32 ]. Considering that about half of HSA-MG protein is capable of binding with MPO ( Figure 3 ), it is not apparent that all subsets of modified albumin inhibit activity of MPO with the same efficacy.…”

“…The function P ( R ) = γ c ( R ) · R is evaluated, where γ c ( R ) is the characteristic function of thickness. The visualization of scattering objects based on SANS data was performed according to the ATSAS manual using DAMMIN [32] as was described earlier [33].…”
