Catabolism of protocatechuate by Bacillus macerans

Abstract: An aerobic endospore-forming bacterium, tentatively identified as a strain (JJlb) of Bacillus macerans, was isolated by enrichment on 4-hydroxybenzoate (4HBA), using as an inoculum soil taken from a 50°C Idaho hot spring. Enzymatic analyses of cells, grown on succinate and 4HBA indicated that strain JJ-lb degrades 4HBA by way of the novel protocatechuate (PCA) 2,3-dioxygenase pathway. Purification of the PCA 2,3-dioxygenase by affinity chromatography allowed the first observation of the immediate ring fission p… Show more

“…These observations were supported by correlating their abundance distribution. The abundances of the film-associated taxa (bins 3, 5, 16,28,32) are significantly positively-correlated to each other. Similarly, the abundances of free-living taxa (1,2,9,10,12,13,14,15,17,21,23,26,30,33) are positively correlated with each other.…”

“…Cell extracts were examined for the presence of the following enzymes: 4,5-dihydroxyphthalate decarboxylase, 3,4-dihydroxyphthalate decarboxylase, protocatechuate 4,5-dioxygenase, protocatechuate 3,4-dioxygenase, and protocatechuate 2,3-dioxygenase. Assays were performed by monitoring disappearance of substrates and appearance of products by UVvisible spectrometry, on the basis of spectral properties reported by others for these enzyme systems (8,11,29,31). Spectral changes at 30°C were recorded with a Perkin-Elmer Lambda 6 spectrophotometer.…”

“…B . macerans JJ-lb degraded protocatechuate via ring cleavage and subsequent enzymatic decarboxylation of the ring fission product (Crawford et al, 1979 ). Initially, protocatechuate-2,3-dioxygenase catalyzes the ring cleavage of protocatechuate to 5-carboxy-2-hydroxymuconic semialdehyde that is further decarboxylated to 2-hydroxymuconic semialdehyde.…”
