Caspase‐dependent cleavage regulates protein levels of Epstein–Barr virus‐derived latent membrane protein 1

Abstract: Epstein-Barr virus (EBV)-encoded latent membrane protein-1 (LMP1) plays pathogenic roles in EBV-related diseases. Thus, host cells employ several mechanisms to regulate LMP1 functions, and we previously reported possible regulation by signal transducing adaptor protein-2 as well as BS69. Here, we found that caspase-3 mainly degraded LMP1 proteins in HeLa cells, leading to decreased NF-jB and STAT3 activation. Caspase-3 cleaved the consensus DNTD sequences in the CTAR3 region of LMP1. Of importance, LMP1 expres… Show more

“…Finally, considering the association of EBV infection in the biology of NPC, the involvement of viral proteins such as latent membrane protein1 (LMP1) in the induction and/or membrane recruitment of pro-caspase-3 can also not be ruled out. In this regard, not only caspase-3 was shown to proteolytically cleave LMP1, but more importantly, LMP1 expression was reported to induce the expression of caspase-3 [ 44 ].…”

“…In addition, the cleavage of other cellular [ 56 , 91 , 92 , 93 , 94 ] or potentially viral proteins by caspase-1 and other caspases could also contribute to EBV reactivation. In addition to caspase cleavage of BPLF1, caspase-3 was reported cleave LMP1 in Hela cells while the functional importance is not clear [ 95 ]. Using bioinformatic tools PeptideCutter and GraBCas [ 96 , 97 ], we also predicted the potential caspase cleavage sites for EBV proteins and found that many other viral proteins, such as BCRF1/vIL10, may also be potentially cleaved by caspases ( S2 Table ).…”

“…Specifically, the median survival for patients with no expression of JAK2 or STAT3 was 58.7 ± 5.3 or 53.6 ± 13.1 months, while those with positive expression of JAK2 or STAT3 had median survival times of 33.6 ± 19.7 or 39.5 ± 20.8 months, respectively [ 274 , 275 ]. In NPC, this pathway is constitutively activated by LMP1 through CTAR3 activation and upregulation of IL-6 [ 276 278 ]. In fact, STAT is activated in a positive feedback loop manner, whereby STAT activation signalling upregulates the expression of LMP1, in turn induces the LMP1-mediated IL-6 production and results in activation of STAT [ 277 ].…”
