Casein phosphopeptide calcium chelation: preparation optimization, in vitro gastrointestinal simulated digestion, and peptide fragment exploration

Ying Wang,
Ruixue Wang,
Huasong Bai
et al.

Abstract: BACKGROUNDThe calcium is closely related to the formation of bones and teeth, cell metabolism and other physiological activities. In this work, casein phosphopeptide‐calcium chelate (CPP‐Ca) was synthesized and the optimal process parameters for the chelation reaction were obtained. The bioavailability of calcium in CPP‐Ca was investigated by in vitro gastrointestinal simulated digestion. The existence of phytic acid and oxalic acid in digestion system was evaluated to clarify the calcium holding ability of CP… Show more

“…In vitro models must be created to investigate the calcium release patterns of different calcium supplements [ 30 ]. Compared to Ca 3 (PO 4 ) 2 , LHWCC was less affected by pH, which was similar to the in vitro digestion results of CPP-Ca [ 31 ]. Jiang et al [ 27 ] found that most peptide calcium chelates were soluble after digestion, but the dialysis rate was significantly lower than the solubility.…”

“…30,34 Consistent with existing literature, numerous casein-derived peptides, including CPPs, were identified following gastrointestinal digestion of dairy products. This has been demonstrated in vitro after SGID, 35–37 in vivo in minipigs 38 or in vivo in human. 20,39 In the current study, we identified 145 casein peptides, including 29 CPPs, after SGID of the CPP-preparation (Fig.…”

“…The team also discerned SNPrs188303909 at the 2q14.2 locus, which was hypothesized to distally regulate the expression of engrailed homeobox 1 (EN1) by modulating DNA methylation and the binding of E2F6. EN1 has been demonstrated to upregulate the expression of the coiled‐coil domain containing 170 (CCDC170) and collectin subfamily member 10 (COLEC10) through binding to osteoporosis‐associated SNPs, which in turn affects bone density and fracture risk (Wang, Wang, et al., 2024). Recently, Chen et al.…”
