Cascade Reactions in Total Synthesis

Abstract: The design and implementation of cascade reactions is a challenging facet of organic chemistry, yet one that can impart striking novelty, elegance, and efficiency to synthetic strategies. The application of cascade reactions to natural products synthesis represents a particularly demanding task, but the results can be both stunning and instructive. This Review highlights selected examples of cascade reactions in total synthesis, with particular emphasis on recent applications therein. The examples discussed he… Show more

“…Although widely used in organic synthesis and catalytic process [86], recently, the cascade reaction is employed as well in polymer mechanochemistry. As a specific form of covalent mechanoluminescence, cascade mechanoluminescence involves multi-steps conversion of mechanical energy to luminescence.…”

“…The most useful MCRs have additional advantages of selectivity, synthetic convergence, atom-economy and green credentials. [4][5][6][7][8][9] Further, MCRs are perfectly amenable for automation to generate combinatorial libraries of biologically relevant heterocyclic scaffolds and hence play a pivotal role in lead identification and drug development programmes.…”

“…One pot tandem reactions, such as the RauhutCurrier reaction, are important for the efficient construction of complex chemical structures and therefore are an active research area in the synthetic community. [23][24][25][26][27][28][29][30][31] The intramolecular Rauhut-Currier reaction 32 has experienced a resurgence based on groundbreaking studies by Krische, 33 Murphy, 34,35 and Roush. 36 Under the influence of catalytic phosphine, conjugated systems can be merged to form a new ring while retaining one conjugated system for subsequent functionalization.…”
