This study reports the protective role of the aqueous extract of Syzygium aromaticum (ESA) against lead (Pb)-induced neurotoxicity in mice. Thirty male mice weighing between 18 g and 25 g were randomly divided into five groups. (1) Group 1 (control group), (2) group 2 (Pb-test group): was administered with a solution containing 0.1% (w/v) of lead acetate (PbAc), (3) group 3 (ESA + Pb100 group): was administered with 0.1% (w/v) of PbAc followed by 100 mg/kg of S. aromaticum extract by gavage, (4) group 4 (ESA + Pb200): was administered with 0.1% (w/v) of PbAc followed by 200 mg/kg of S. aromaticum extract, and (5) group 5 (ESA-group): was administered with 100 mg/kg of S. aromaticum. Level of lead was determined by atomic absorption spectroscopy. Cerebral cortex synaptosomes prepared from mice administered orally with lead-acetate shown a significantly increased (p < .05) in tyrosine hydroxylase and protein carbonyl level and significantly decreased (p < .05) superoxide dismutase, glutathione reductase, and glutathione transferase activities. Also, there was a significant increase in brain lead concentration level, however, it was observed that S. aromaticum significantly reduced (p < .05) the level of lead at all tested doses. S. aromaticum rescued cerebral cortex synaptosomes from lead-induced neurotoxicity by relieving oxidative stress and abating elevated tyrosine hydroxylase activity. Moreover, S. aromaticum at the different dose grade (100 mg and 200 mg) abrogated the loss of motor performance in mice groups induced with lead. Altogether, our findings showed that S. aromaticum possesses antioxidant and neuro-modulatory potential against lead-induced neuronal damage. Practical applicationsEnvironmental pollution with heavy metals is a known public health concern and their incremental concentrations in soil and water have risen to an unprecedented degree.Lead is one of the top 10 contaminants on the WHO's list of substances of greatest public health concern that impact the brain. Amikacin (AK) is frequently used in the treatment of gram-negative and some gram-positive infections. However, its use is limited due to nephrotoxicity due to the increase in reactive oxygen radicals. The aim of this study was to investigate the role of carvacrol (CAR) against AK-induced nephrotoxicity in rats. Thirty-two Sprague Dawley rats were randomly divided into four groups as control (Vehicle), AK (400 mg/kg), CAR + AK (80 mg/kg CAR + 400 mg/kg AK), and AK + CAR (400 mg/kg AK + 80 mg/kg CAR) groups. AK and CAR were administered via intramuscular and per-oral for 7 days, respectively. Blood and kidney tissue samples were taken at the end of the experiment. Renal function and histopathological changes were compared, and the relevant parameters of oxidative stress and inflammation were detected. Histopathological findings (necrotic changes and dilatation and inflammatory cell infiltration) significantly increased in the AK group compared to the control group. Also, the rats in the AK group lost weight significantly. It was found that CAR treatment before and after AK significantly improved nephrotoxicity histopathologically (p < 0.05). However, this improvement was not detected biochemically. These results show that CAR treatment before and after AK improves nephrotoxicity in the histopathological level.

The Neuroprotective effect of carvacrol, which has anti-inflammatory and antioxidant effects, on infarcted cerebral tissue is present in literature, but this contribution was not sufficiently clarified in terms of biochemistry. It is aimed to investigate the effect of orally administered carvacrol on plasma and intraparenchymal levels of TBARS, GSH, SOD, CAT, GSH-Px, IL-1β, IL-4, and TNF-α after the formation of global ischemia in cerebral tissue. Four groups were formed, each containing ten Wistar albino rats. After anesthesia and analgesia, bilateral carotid communis arteries of rats in the first two groups were clamped for 15 minutes with aneurysm clips. Oral 50 mg/kg/day carvacrol was administered for 15 days to the first group (I/R+CRV) of these two groups in which cerebral ischemia-reperfusion (I/R) was established. On the other hand, %0,01 carboxymethylcellulose (CMC), which is a solvent of carvacrol, at a same volume of first group was administered orally for the same duration to the other group in which also I/R was established (I/R+CMC). In the other two groups in which ischemia was not induced, only carotid artery dissections were made and sutured again. In these two groups, 50 mg/kg/day of carvacrol was administered to the first group (CRV). The Same dose of CMC was administered to the second group (control group). After all these treatments, plasma was collected, and brain tissue was dissected from all groups at the end of the 15th day. Carvacrol can be included in the possible treatment regimen of cerebral stroke with the help of other studies that can be supported on this topic.
