Cartilage homeostasis in health and rheumatic diseases

Abstract: As the cellular component of articular cartilage, chondrocytes are responsible for maintaining in a low-turnover state the unique composition and organization of the matrix that was determined during embryonic and postnatal development. In joint diseases, cartilage homeostasis is disrupted by mechanisms that are driven by combinations of biological mediators that vary according to the disease process, including contributions from other joint tissues. In osteoarthritis (OA), biomechanical stimuli predominate wi… Show more

“…We examined the effects of systemic WNT-974 exposure on two other tissues that are reliant upon Wnt signaling for regeneration: hair follicles and cartilage (13,14). In both tissue types, suppression of Wnt signaling, following either depilation or the introduction of puncture wounds in the ear, resulted in delayed hair and cartilage regrowth ( Fig.…”

“…At the cellular level, chondrocytes respond to this cytokine via a cascade of events leading to loss of balance between cartilage anabolism and catabolism, associated with phenotypic modulation and activation of the normally quiescent chondrocytes (Goldring and Marcu 2009;Goldring et al 2008). In OA cartilage, IL1b and TNFa co-localize with increased levels of matrix metalloproteinases (MMPs) (Tetlow et al 2001), and IL1b was shown to upregulate MMP expression levels in normal human chondrocytes, OA chondrocytes, and chondrosarcoma cells (Tetlow et al 2001;Fan et al 2005;Mix et al 2001).…”

“…These unique properties are due to the structural organization of the main macromolecules that compose the cartilage extracellular matrix, namely collagens and proteoglycans. Chondrocytes, the only cell type present in articular cartilage, are embedded in the extracellular matrix and are responsible for maintaining its homeostasis by ensuring the synthesis and turnover of its components [Martel-Pelletier et al, 2008;Goldring and Marcu, 2009].…”
