Carrier and Hapten Functions in Immune Deviation

Abstract: The effects of the intravenous injection of DNP-Ficoll on delayed hypersensitivity reactions to bovine gamma globulins (BGG) and anaphylactic reactions to the hapten induced by immunization with DNP-BGG in Freund’s complete adjuvant were studied in guinea pigs. Animals treated with DNP-Ficoll exhibited stronger delayed hypersensitivity to BGG and lower anaphylactic reactions to the hapten than control animals treated with saline, Ficoll alone or with an unrelated protein antigen. These results could be explain… Show more

“…In the guinea pig, helper T cells have been already functionally dis sociated from T cells involved in delayed hypersensi tivity [Neveu and Borduas, 1974;Silver and Benacerraf, 1974], Similarly, T cells involved in delayed hy persensitivity and T cells involved in contact dermati tis could belong to distinct T cell subclasses [Neveu, 1979].…”

“…Antigenic competition could take place at the level of T cells responsible for both cellular immu nity and helper function in antibody response. How ever, helper T cells and T cells involved in cellular im munity are not necessarily the same cells: their func tions have been dissociated in guinea pigs [16,25], and these two cell populations have been separated by physical means [8] and express distinct phenotypes in mice [11], The enhanced DH reactions to the carrier after the injection of the hapten coupled to an unre lated carrier, as seen in experiment 1 (table I), could have resulted from a nonspecific adjuvant action of T cells triggered by either the unrelated carrier [13] or haptenic determinant of the hapten-protein carrier conjugate [9]. These possibilities have been excluded in experiment 4 (table II), where antigenic competi tion was observed after treating the animals with the hapten coupled to a T-independent carrier.…”
