Cardiorenal Effects of Long‐Term Phosphodiesterase V Inhibition in Pre–Heart Failure

Abstract: Background Phosphodiesterase V (PDEV) is upregulated in heart failure, leading to increased degradation of cGMP and impaired natriuresis. PDEV inhibition improves the renal response to B‐type natriuretic peptide in animal models. We tested the hypothesis that long‐term PDEV inhibition would improve renal function and cardiorenal response after short‐term volume load in subjects with pre–heart failure. Methods and Results A total of 20 sub… Show more

“…Each cluster then applies a local general linear model, and the K is adjusted to maximize the R2 for all local models. LIME-SUP [31] replaces the unsupervised clustering technique used in K-LIME with a supervised partitioning tree to enhance the quality of explanations. NormLIME [32] introduces a class-specific global explanation by aggregating and normalizing a group of defined local explanations.…”

“…This is due to the distinctive temporal association between attributes in time series data, which must be considered to produce valid and substantial explanations. Signal data exhibits intricate temporal dependencies and unique characteristics, making it challenging to perturb the Another substantial limitation impacting the credibility of LIME explanations stems from the fundamental linearity assumption inherent in the methodology [31]. LIME operates under the premise that the behavior of a machine learning model can be approximated by a locally linear model.…”

“…Considering the impact of reduced NO bioavailability on the pathophysiology of CVDs, strategies aimed at compensating for this deficit may yield benefits for cardiovascular health. Some of these strategies focus on the NO canonical pathway, involving the modulation of NOS action and the inhibition of cGMP degradation [ 100 , 101 ]. However, the enterosalivary cycle of nitrate is known to serve as an additional source of NO, complementing the canonical pathway.…”
