Cardioactive Diterpenoids from the Roots ofSalvia amplexicaulis

Abstract: Five diterpenoids, three steroids, four triterpenoids and one flavonoid were isolated from the roots of Salvia amplexicaulis Lam. (Lamiaceae). Structures of these compounds were elucidated by spectroscopic analysis. The crude extract and the pure compounds were tested for cardiovascular parameters using Wistar Albino rats. The crude extract, and 7-oxo-abieta-9,12,14-triene, ferruginol, stigmast-4-en-3-one showed a vasodepressor effect.

“…Several biological activities have been reported for ferruginol, including antimicrobial effects (Politi et al 2003), inhibitory effects on Epstein-Barr virus early antigen activation induced by 12-O-tetradecanoylphorbol 13-acetate (Iwamoto et al 2003), cardioactive activity (Kolak et al 2001;Ulubelen et al 2002) and hypotensive effects (Ulubelen et al 2000).…”

“…The compounds 1, 2, 3, 6 and 20 have been previously reported from natural sources (Ying & Kubo 1991). Ferruginol was previously reported as a constituent of the resin of Podocarpus ferrugineus, Podocarpus totara, Dacrydium spp., Cupressus spp., Cryptomeria japonica, Salvia syriaca and the roots of Inula royleana (Dictionary of Natural Products on CD ROM 2005), Cephalotaxus harringtonia (Politi et al 2003), Thuja standishii (Iwamoto et al 2003), Salvia eriophora (Ulubelen et al 2002), Salvia amplexicaulis (Kolak et al 2001) and Salvia syriaca (Ulubelen et al 2000). Compound 9 was previously described in a patent for photosensitive material (Shida et al 1996), while the benzoate 10 was prepared by King et al (1957) to compare synthetic and natural ferruginol as crystalline benzoates .…”

“…The acetone extracts of Salvia lanigra and Salvia splendens and the n-butanol extract of Salvia lanigra were repeatedly chromatographed over normal-phase silica gel, silica gel for flash and reversed-phase silica gel to give five diterpenes (1-5), two triterpenioc acids (6 and 7), one sterol (8), two flavones (9 and 10) and one phenolic acid (11). The known compounds were identified as horminone (1) [10][11][12] and 7-O-ethylhorminone (2), [10][11][12][13] ursolic acid (6) and oleanolic acid (7), [14] β-sitosterol (8), [15,16] salvigenin (9) and apigenin (10) by comparing their spectral data (mass spectrometry, 1 H-and 13 C NMR) with those reported previously. [17][18][19] Cell survival was evaluated by adding 10 μL of 5 mg/ml MTT in 0.1 mM, pH 7.4 phosphate buffered saline to each well, and re-incubating plates in 5% CO 2 /air for 4 hrs at 37°C.…”

“…This indicated that carbon C-11 was therefore substituted by a hydroxy group, as deduced from its ESIMS [18,26]. The chemical shifts at δ assumed [11,12,[20][21][22][23][24]. Complete assignment of protons and carbons of 2 (Table 1) ) and two carbonyls.…”
