Cardiac H2S Generation Is Reduced in Ageing Diabetic Mice

Abstract: Aims. To examine whether hydrogen sulfide (H2S) generation changed in ageing diabetic mouse hearts. Results. Compared to mice that were fed tap water only, mice that were fed 30% fructose solution for 15 months exhibited typical characteristics of a severe diabetic phenotype with cardiac hypertrophy, fibrosis, and dysfunction. H2S levels in plasma, heart tissues, and urine were significantly reduced in these mice as compared to those in controls. The expression of the H2S-generating enzymes, cystathionine γ-ly… Show more

“…The H 2 S levels in the heart tissues and bone marrow-derived macrophages from WT mice and KO mice was measured as previously described with minor revision (17). In brief, 30 µL supernatant of heart tissues or BMMs were homogenized in ice-cold Tris-HCl (100 mmol/L, pH 8.5), and 70 µL monobromobimane reagent was added, vortexed away from light, and then added 10 µL 20% Formic acid, following by centrifugation at 12,000 g, 4°C for 20 min.…”

“…High glucose induces cardiovascular dysfunction by activating the PKC/diacylglycerol pathway, calcium dysregulation, inducing endoplasmic reticulum stress, and deregulating autophagy and apoptosis, which have been shown to be mitigated by H 2 S supplementation (9,80,89,156,164). Furthermore, plasma H 2 S levels are decreased in type 2 diabetic patients, high-fat diet-treated mice (type 2 diabetic patients), and streptozotocin-treated rats (9,64,66). The decrease in H 2 S levels in diabetics and the ability of exogenous H 2 S treatment to ameliorate pathological conditions induced by type 1 and type 2 diabetes suggest that H 2 S may be a potential novel treatment option for mitigating cardiomyopathy in diabetics.…”

“…Though, there are a lot of excellent reviews which expound potential physiological function of H 2 S in seed germination, plant growth and development, as well as the acquisition of stress tolerance (Li, 2013; Calderwood and Kopriva, 2014; Hancock and Whiteman, 2014, 2016; Jin and Pei, 2015; Guo et al, 2016; Scuffi et al, 2016; Yamasaki and Cohen, 2016), the role of H 2 S as a candidate signal molecule in plant cross-adaptation was not summarized in depth. Therefore, in this review, H 2 S homeostasis in plant cells under normal growth conditions, H 2 S signaling triggered by adverse environment and H 2 S-induced cross-adaptation to various abiotic stresses are summarized, which further uncovers that H 2 S may be a candidate signal molecule in plant cross-adaptation.…”
