Cardiac ankyrin repeat protein contributes to dilated cardiomyopathy and heart failure

Abstract: Cardiac ankyrin repeat protein (CARP) is a cardiac‐specific stress‐response protein which exerts diverse effects to modulate cardiac remodeling in response to pathological stimuli. We examined the role of CARP in postnatal cardiac development and function under basal conditions in mice. Transgenic mice that selectively overexpressed CARP in heart (CARP Tg) exhibited dilated cardiac chambers, impaired heart function, and cardiac fibrosis as assessed by echocardiography and histological staining. Furthermore, th… Show more

“…Left ventricle internal diameters at end-diastole and end-systole (LVIDd and LVIDs, respectively) were measured. The LV ejection fraction (EF), fractional shortening (FS), left ventricular end-diastolic volume (LVEDV), and left ventricular end-systolic volume (LVESV) were analyzed independently by three investigators who were blinded to the experimental design ( 27 ).…”

“…This state is defined by the expression of cardiomyocyte stress factors including Ankrd1 , Ankrd23 and Xirp2 [Figure 3E & Suppl Fig S4B] . Ankrd1 and Ankrd23 are stress-inducible ankyrin repeat proteins which are elevated in dilated cardiomyopathies 26,27 . Xirp2 encodes cardiomyopathy-associated protein 3 and is upregulated in CMs in response to stress 28,29 .…”

“…[29][30][31] Furthermore, it has been shown that downregulation of CARP expression induces upregulation of ryanodine receptor 2 expression, resulting in slow and continuous entry of Ca 2+ into the cytoplasm. 32 In addition, 2 (CASQ2) is the major Ca 2 +-binding protein on the sarcoplasmic reticulum and the primary Ca 2+ reservoir for the myocardium. Myocardial CARP specifically binds and interacts with specific CASQ2 domains through the CasQ-binding sequence.…”
