Carboplatin and teniposide concurrent with radiotherapy in patients with glioblastoma multiforme

Abstract: METHODS. Fifty-six RESULTS. All 56 patients were evaluated for time to progression (TTP) and median survival time (MST). The TTP was 7.5 months and the MST was 12.5 months. 2 Department of Radiotherapy, Azienda OspedaToxicity manifested as thrombocytopenia and in most cases was acceptable. Four liera, Padova, Italy. patients (7.1%) had radiation necrosis at 2, 2, 7, and 9 months, respectively, from 3 Department of Neurosurgery, Azienda Ospedathe end of radiotherapy. liera, Padova, Italy. “…It is documented in two studies of the synopsis summarised in Table 1 and ranges between 15.4 and 28% and is 29% in our study (Brandes et al, 1998;Beauchesne et al, 1999). Concerning median survival, Gruber et al (1998) achieved 19.2 months with the single agent carboplatinum.…”

“…Our survival and TTP results range in the upper level of those achieved in the limited number of available first-line treatment strategies, which are summarized in Table 1. Therein, the median TTP is 7 (range: 4.2 -9.5) months and the median survival is 12 (range: 9 -19.2) months (Table 1; Fetell et al, 1997;Brandes et al, 1998;Friedman et al, 1998;Gruber et al, 1998;Beauchesne et al, 1999;Frenay et al, 2000;Grossman et al, 2000;Stupp et al, 2000).…”

“…Radiation-and chemotherapy-induced necrosis occurs slowly, usually several days after treatment (Watterson et al, 1993;Hruban et al, 1989;Minchin et al, 1988;Brandes et al, 1998;Packer et al, 1994;Powers et al, 1991). Upregulation of the Bax gene induced by radiation and chemotherapy, combined with mitochondrial damage due to hypoxia from vascular damage, may induce tissue necrosis by the mechanism reported here.…”

“…A recent phase-II study of intensified chemotherapy, where BCNU was preceded by carboplatin and teniposide, administered during radiotherapy to 56 patients with glioblastoma multiforme, resulted in a median time to progression of 7.5 months and a median survival of 12.5 months [10]. Comparable figures have been achieved in various single agent trials [54].…”
