Carbon Nanotube-Delivered Ic9 Suicide Gene Therapy for Killing Breast Cancer Cells In Vitro

Abstract: Aim: To induce a safe death to MCF-7 human breast cancer cell line through gene therapy based on iC9 suicide gene. Materials & methods: To induce apoptosis to MCF-7 cell line, iC9 gene was transfected using pyridine-functionalized multi-walled carbon nanotubes. Then, to enhance chemotherapy, iC9 suicide gene therapy was performed alongside. Results: The results show that the MCF-7 cells were efficiently eliminated in a high percentage by this approach. Furthermore, the suicide gene by itself/in combination… Show more

“…Recent research has shown that this strategy is successful in treating chemo-resistant cancer cell lines [99] and also enhances the effectiveness of radiation during therapy [100]. In this context, Dargah et al demonstrated that iC9 gene induces apoptosis in MCF-7 human breast cancer cell lines [101]. In this study, pyridine modified MWCNTs were used as carriers to transfer the gene of interest.…”

“…A combination of the introduced pf-MWCNTdelivered iC9 suicide gene therapy and chemotherapy was also performed. Reports showed that pyridine-functionalized multi-walled carbon nanotubes (pf-MWCNT)-delivered iC9 suicide gene therapy and is an effective strategy for killing cancer cells; combination therapy works even better than monotherapy due to overcoming cell cycle arrest resulting from chemical drugs [100]. A novel photoactivatable RNAi system was designed for targeted gene expression in tumor cells.…”

“…Another gene that has been used is the Escherichia coli cytosine deaminase gene; it encodes for an enzyme that catalyzes conversion of 5-fluorocytosine (5-FC) into 5-fluoruracile (5-FU), a drug used in conventional chemotherapy of HCC, prostate, colon, and breast cancers because it causes cell proliferation inhibition and induces cell death (14,67,71). Furthermore, this approach has been carried out by using a suicide gene which encodes for a modified human Caspase 9 (iCas9), which is strongly recognized by the bioinert synthetic molecule (B/B Homodimer, AP20187), alone or delivered by mesenchymal stromal cells (MSCs), inducing a dimer formation and activation apoptosis pathway in breast and lung cancer cells (72)(73)(74).…”
