Carbamylcholine‐ and 5‐hydroxytryptamine‐induced contraction in rat isolated airways: inhibition by calcitonin gene‐related peptide

Abstract: 1 The effects of rat and human x-calcitonin gene-related peptide (a-CGRP) were investigated in isolated smooth muscle preparations obtained from three levels of the rat respiratory tract.2 Neither peptide (10--10-16M) had any effect on resting tension or on carbamylcholine (10-6M)-induced tone of trachea or main bronchus. In contrast, CGRP sometimes reduced spontaneous or carbamylcholine-induced tone of lung parenchymal strips. 3 CGRP produced a significant rightward shift of the log concentration-response cur… Show more

“…In other animal species, CGRP has been shown to have no contractile effect on the airway smooth muscle in the rabbit (26), pig (27,28), rat (10,20), and mouse (29) in vitro and in the sheep in vivo (30). Finally, CGRP has been noted to cause relaxations in pig trachea (27) and bronchi (28) precontracted with carbamylcholine and histamine, respectively, and in mouse airways precontracted with carbachol (29).…”

“…The inhibitory effect of CGRP on SP-induced contractions was investigated essentially as described previously (10). Each tissue preparation was first primed with a concentration of carbamylcholine (10 Ϫ 6 M) to evaluate its responsiveness.…”

“…(Tschirhart et al, 1990) and in the human bronchus (Palmer et al, 1987) in vitro. On the other hand, CGRP is shown to have no effect on the airway smooth muscle in the guinea-pig (Bhogal et al, 1994) and pig trachea (Kannan & Johnson, 1991) in vitro and in the sheep in vivo (Parsons et al, 1992), and to cause inhibition of the carbachol-and 5-hydroxytryptamine-induced contraction of rat isolated airways (Cadieux et al, 1990). Furthermore, CGRP is reported to cause relaxations in guinea-pig trachea precontracted with KCl and PGF2a (Bhogal et al, 1994) and in pig trachea (Kannan & Johnson, 1992) and mouse bronchus (Manzini, 1992) precontracted with carbachol.…”

“…CGRP modulates contraction induced by 5-HT or carbachol in the airway [7] and the actions of vasodilators [3]. It has been reported that in airway epithelia CGRP modulates ciliary beat frequency and mucus secretion [33] and causes proliferation of epithelial cells [37].…”
