Capture enzyme-linked immunosorbent assay to detect specific immunoglobulin E in sera of patients with paracoccidioidomycosis.

Abstract: Abstract. Paracoccidioidomycosis (PCM) is the most frequent systemic mycosis in South America. The disease is characterized by a polyclonal activation of B cells, resulting in hyperimmunoglobulinemia. The production of immunoglobulin (Ig) E in deep mycosis has been related to the severity of the disease. However, the detection of specific IgE in sera of patients is difficult because of the competition with the IgG. We compared a capture and an indirect enzyme-linked immunosorbent assay (ELISA) technique to det… Show more

“…Dotted line represents the cut-off value, which is the mean value plus 3·SD of antibody titers of healthy individuals in their twenties and thirties in a malaria-free area IgM. Antibody capture ELISA is known to be a better method for determining the specific isotype, avoiding competition from other isotype antibodies and has been used in IgE detection assay in parasite disease (Mamoni et al 2001). By using an antibody capture assay, the minor isotype antibodies against PvMSP1c could be determined (Fig.…”

“…83 Several other immunologic assays, particularly immunodiffusion, are valuable diagnostic adjuncts. 84,85 The intradermal reaction ( paracoccidioidine) is not reliable for diagnosis in endemic areas. 59,74,85 Slow-acting sulfonamides are the drugs of choice in the treatment of PM.…”

“…However, due to the time and material required to perform these techniques, serologic tests have been developed to detect both antibodies (7,19,30,31) and antigens (14,15,16,22). Serologic tests are important tools when other procedures are not available as well as for posttherapy follow-up (21).…”
