BackgroundAllium sativum L. (A.S.) “garlic”, one of the most interesting medicinal plants, has been suggested to contain compounds that could be beneficial in numerous pathological situations including cancer. In this work, we aimed to assess the immunomodulatory effect of A.S. preparation on human peripheral blood mononuclear cells from healthy individuals.MethodsNontoxic doses of A.S. were identified using MTT assay. Effects on CD4+ or CD8+ T lymphocyte proliferation were studied using flow cytometry. The effect of A.S. on cytokine gene expression was studied using qRT-PCR. Finally, qualitative analysis of A.S. was performed by HPLC approach. Data were analyzed statistically by one-way ANOVA test.ResultsThe nontoxic doses of A.S. preparation did not affect neither spontaneous nor TCR-mediated CD4+ or CD8+ T lymphocyte proliferation. Interestingly, A.S. exhibited a statistically significant regulation of IL-17 gene expression, a cytokine involved in several inflammatory and autoimmune diseases. In contrast, the expression of IL-4, an anti-inflammatory cytokine, was unaffected. Qualitative analysis of A.S. ethanol preparation indicated the presence of three polyphenol bioactive compounds, which are catechin, vanillic acid and ferulic acid.ConclusionThe specific inhibition of the pro-inflammatory cytokine, IL-17 without affecting cell proliferation in human PBMCs by the Allium sativum L. preparation suggests a potential valuable effect of the compounds present in this plant for the treatment of inflammatory diseases and cancer, where IL-17 is highly expressed. The leaves of the Capparisspinosa were collected from the eastern Radwaniya/Baghdad duringthe months April, May, June, July, September, and September. The leaves were extracted by water and alcohol. The effectiveness of anti-tumor and anti-leishmania for the water and alcohol extracts of al-Qabar leaf samples for May and September, which were characterized by the highest percentage of active compounds for the rest of the tested months, was evaluated by adopting the results of a previous study for the same researchers or chemical composition and leaf content of active compounds The concentrations of 250, 500, 1000 mg.L 1 were evaluated for anti-tumor efficacy. The anti-leishmania efficacy was tested at 250, 500 and 1000 mg L 1. The sample of the alcohol extract for May collected complex showed the highest anti-tumor activity at 63.5% at 500 mg.L 1 and the highest efficacy of the samples of alcohol extract for September was 56.1% at the concentration of 250 mg.L 1 , while the water extract for the May sample did not show any effectiveness even at the highest concentration 1000 mg L 1 , but the September collected sample was effective at 49.3% In the case of the inhibitory efficacy of leishmania, the alcohol extract of the May sample did not show on inhibitory activity at the lowest concentration. The effectiveness of 41.3% was observed at 500 mg.L 1 , but decreased to 14.8% at the highest concentration of 1000 mg L-1. Of the same month at the lowest concentration of 37.8% and decreased effectiveness there was an increase in the anti-leishmania effect of the September extract of the sample with increased concentration. There was of its effectiveness at the lowest concentration of a user and then reached 8 and 37% at the concentration of 500 and 1000 mg.L 1 with the same percentage, and the water extract for the September sample did not show any efficacy at the minimum and highest concentrations, except the concentration of 500 mg.L 1 , which reached 11%.

Background: Orofacial pain is a form of inflammatory pain usually treated with corticosteroids, which have many side effects. Objective: The aim of this study was to investigate the clinical and histological profile of antinociceptive effects of hydroalcoholic extract of caper (Capparis spinosa) for the first time in the orofacial region. Methods: This experimental study was carried out at Babol University of Medical Sciences in 2018. Thirty-six male Wistar mice were divided into 6 groups: The first group received saline, the second group received dexamethazone, and four groups received different doses (10, 20, 50, 100 mg/kg) of caper extract. In the formalin test, 1% formalin solution was injected into the right submucosal layer of the lip and lateral area of the nose. Pain intensities were recorded at 5-min blocks for 60 min after injection. Dose effect of caper on pain was recorded. The mice were euthanized and the oral area was biopsied and stained with Hematoxylin-eosin, Toluidine blue, and Congo red. Data were analyzed using repeated measures ANOVA and t-test by IBM-SPSS version 20. Results: Caper produced antinociceptive effects in comparison with saline groups (p<0.001). Dose effect on pain was significant (p<0.001). The highest antinociceptive effect was observed in the caper group receiving 100 mg/kg of agent 15-20 minutes after injection. The highest pain level was observed in the group that received 20 mg/kg of caper (p<0.001). Dexamethazone antinociceptive effect was greater than that of the saline and the dose of 20 mg/kg of caper (p<0.001). Antinociceptive effects in two groups (100 mg caper and dexamethazone) were equal (p>0.999). Histopathologic examination revealed the highest thickness of epithelium, fibrous, and muscular tissue density and the lowest inflammatory infiltration at the dose of 100 mg/kg of caper. Conclusion:The results of the present study suggest that hydroalcoholic extract of caper possesses antinociceptive activity in a dose-dependent manner and caper-induced antinociception might be mediated, at least in part, by anti-inflammatory effects.
