Capillary electrophoresis with field-enhanced stacking for rapid and sensitive determination of strychnine and brucine

Abstract: A new capillary electrophoresis procedure with field-enhanced stacking concentration for the analysis of strychnine and brucine is established. After optimization of the separation and concentration conditions, the two alkaloids can be separated within 5 min and quantified with high sensitivity (The detection limits were 1.0 ng mL(-1) for strychnine and 1.4 ng mL(-1) for brucine). The method was useful for qualitative and quantitative analysis of strychnine and brucine in Strychnos nux-vomica L with recovery o… Show more

“…For example, CE has low detection sensitivity due to the small sample volume. [8][9][10][11] Recently, various on-column pre-concentration techniques such as pH-mediated base stacking, 4,5 micellar electrokinetic chromatography (MEKC), 6 field amplification or enhancement, [9][10][11] moving reaction boundary (MRB), 12 micelle to solvent stacking (MSS) 13 and base stacking (BS) 14,15 have been developed and applied to eliminate these constraints. Some methods combine two concentration techniques, such as field-enhanced sample injection (FESI) and sweeping for the on-line concentration of analyte.…”

“…Hu et al employed FASS to the on-line concentration of the alkaloids from Sophora flavescens [8]. With field-enhanced stacking Hu et al further established an analysis of strychnine and brucine in Strychnos nux-vomica [9]. Pospíšilová et al separated and determined five flavonoids in Hypericum perforatum by capillary zone electrophoresis (CZE) with isotachophoretic (ITP) sample pre-treatment using on-line column coupling configuration [10].…”

“…A large number of applications of sample stacking in environmental and biomedical analysis have been reported [6]. In the filed of phytopharmaceutical or natural product analysis, not a few studies concerning on-column concentration have also been carried out in recent years [7][8][9][10][11][12][13]. Unger and Stöckigt applied CE with field amplified sample injection (FASI) to detect the alkaloids in crude extracts from roots of Berberis vulgaris and Hydrastis Canadensis [7].…”

“…FASI has been applied in capillary-and microchip-based electrophoresis for the analysis of various chemically charged species, including pharmaceutical compounds, inorganic ions, amino acids, peptides and proteins [15][16][17][24][25][26][27][28][29]. Electro-kinetic stacking of neutral analytes in MEKC has also been reported [30].…”
