Cannabinoid-Elicited Conditioned Place Preference in a Modified Behavioral Paradigm

Abstract: Cannabinoids are the active ingredients in marijuana, which is among the most widely used addictive drugs despite the well-documented harmfulness related to its abuse. The mechanism underlying cannabinoid addiction remains unclear, which is attributed partially to the difficulty in behavioral testing of high-dose cannabinoids using the conditioned place preference (CPP) model. Here, we optimized conditions for establishing CPP with the synthetic cannabinoid HU210 intraperitoneally administered at a high dose. … Show more

“…Protein levels were measured using western blot analysis, with β‐actin used to ensure equal loading of the samples, according to previously published details , and in Supporting Information Material and Methods 1. Primary antibody dilutions used were as follows; SYS, SYP, BDNF, CamK‐II, CamK‐IV, Calcineurin A (Abcam, USA), ERK1/2, p‐ERK1/2 (Cell Signaling Technology, MA, USA), and β‐actin (Santa Cruz).…”

“…Interestingly, as previously demonstrated with Δ 9 -THC, prior exposure to Δ 9 -THC was required to “unmask” rewarding effects of JWH-018 [94] and pre-exposure to HU-210 prior to CPP conditioning sessions was necessary to obtain place preference with HU-210. [95]. At the time of this writing, very few SCBs have been tested in CPP experiments.…”
