Canine mammary carcinoma cell line are resistant to chemosensitizers: verapamil and cyclosporin A

Abstract: Cancer chemotherapy can fail in many ways. One of the most significant is the development of multiple drug resistance (MDR), which constitutes a serious clinical problem. The development of MDR relates to the expression of a major membrane pump, P-glycoprotein (P-gp). Thus, currently one of the goals of experimental and clinical oncology is to decrease its activity. So far, many different P-gp inhibitors are available, but their efficacy is still questionable and requires further study. The aim of our study was… Show more

“…Drug resistance to vinblastine in CMTs cell lines is documented [30] and a high expression of P-gp [29] and BCRP [28] in CMT cell lines led to the conclusion that the use of DOX could lead to chemoresistance in treated bitches [28].…”

“…P-gp and BCRP expression in CMTs has been demonstrated using techniques able to detect their presence at the subcellular level [22][23][24][25][26][27], however studies investigating the functionality of the pumps with regard to the chemotherapeutic exposure are still incipient in the dog [28][29][30].…”

“…Król et al (2014) treated with vinblastine two canine mammary tumors cell lines (CMT-W1 and CMT-W2) and two cell lines isolated from their lung metastases (CMT-W1M and CMT-W2M) in order to evaluate the effect of P-gp inhibitors verapamil and cyclosporine A and their possible implication in multidrug resistance. The results from qRT-PCR revealed that the multidrug-resistance gene (MDR1) known to be involved in P-gp synthesis was overexpressed in all canine mammary cell lines [ 141 ].…”

“…Chemotherapeutic molecules like anthracycline are substrates of many ABC transporters, and the onset of chemoresistance to these compounds has been attributed to the expression of P-gp and BCRP in human oncology. 14,45,47 Some studies on canine mammary tumors have investigated P-gp and BCRP expression, 4,16,25,28,32,33,49,53 but the role of these chemoresistance modulators is still incipient. A previous study designed and carried out by some of the present study’s authors (M.L., B.B., G.S., C.B.)…”
