Cancer Stem Cells: Novel Target Using Dietary Components for Prevention and Treatment

Abstract: Cancer is the second leading cause of mortality in the United States and no significant treatment is currently available. Although an increasing number of therapeutic options exist for patients with advanced disease, their efficacy is time limited and non-curative. Presently approximately close to 60% of cancer patients in the United States are believed to utilize therapies derived from plants, herbs, flowers, or nutrients either exclusively or concurrently with traditional chemotherapy or radiation therapy. A… Show more

“…We have recently demonstrated that Doublecortin Calmodulin-like Kinase 1 (DCKL1) is a marker of quiescent stem cells in a variety of cancers including colon cancers [32]. Since, BMW extracts inhibited colonosphere formation, we determined whether DCLK1 expression was affected.…”

“…Spheroids were prepared as previously described [ 5 ]. Briefly, 1000 cells/well in DMEM/F12 (Invitrogen, France) supplemented with EGF (20 ng/ml, Invitrogen) and B27 supplement (1x, Invitrogen) were distributed in in ultra-low attachment 6 well plates.…”

“…Recent evidence suggests the existence of small populations of cancer initiating cells (CSC), which are believed to be responsible for tumor initiation and progression as well as resistance to chemotherapy and radiation. Pancreatic CSC can be allowed to divide and grow in ultra-low binding tissue culture dishes to form multicellular spheroids called pancospheres [ 5 , 6 ]. Recent studies have identified a rare group of label retaining cancer stem cells that have the capacity to regenerate and also divide and develop tumors.…”

“…Natural products or compounds derived from natural products have been included in the most effective cancer treatments to date. Even today, some natural compounds are known to have antioxidant, anti-proliferative, and pro-apoptotic effects in many cancers such as breast, colon, brain, melanoma, and pancreas [7].…”
