Objective: there is an urgent need to identify molecular biomarkers rather than clinical markers to distinguish aggressive prostate cancer from the indolent majority for proper treatment and accurate prediction of prognosis. We aimed to investigate the immunohistochemical expression of epithelial-mesenchymal transition (EMt)-related molecules (twist-1 and E-cadherin) and the stem cell marker EZH2 in prostate cancer and to assess their ability to identify the high-risk patients, in a trial to explore their prognostic implications. Material and Method:Immunohistochemical expression of twist-1, E-cadherin and EZH2 in 50 specimens of prostate cancer and 20 cases of benign prostatic hyperplasia were studied. the relationship between their expression and the clinicopathological variables, biochemical recurrence, and biochemical progression-free survival were investigated. Results:Our results revealed that high twist-1, as well as high EZH2 expression, was strongly associated with higher pre-treatment PSa level, gleason score ≥7, advanced tumor stage, lymph node involvement, distant metastasis and biochemical progression. aberrant E-cadherin expression was significantly associated with higher pre-treatment PSa level, gleason score ≥7, advanced tumor stage, lymph node involvement, and distant metastasis. a significant positive correlation between twist-1 and EZH2 expression was found (p<0.001), while E-cadherin expression showed a negative correlation with both markers (p<0.001). a significant association was found between high twist-1, high EZH2& aberrant E-cadherin expression, and shorter biochemical progression-free survival.Conclusion: the high twist-1 expression, aberrant E-cadherin and high EZH2 expression in primary prostate cancer are considered as adverse prognostic markers of an aggressive tumor with high metastatic potential. assessment of their expression level would contribute to the accurate prediction of biochemical progression.

Objective: Cervical cancer has an increasing incidence in developing countries with a predominance of squamous cell carcinoma. In this work, we aimed to analyze the role of EZH2, Endothelin-1, and CD34 as indicators of the aggressiveness in cervical squamous cell carcinoma. Material and Method:Immunohistochemical expression of EZH2, Endothelin-1, and CD34 was studied in 54 paraffin-embedded tissue specimens of cervical squamous cell carcinoma. Their correlation to the clinicopathologic features and the potential angiogenic role were analyzed.Results: High EZH2 expression was noted in 78% of cervical squamous cell carcinoma with a significant relation with tumor grade, FIGO stage and lymph node metastasis (p=<0.001, p=0.007, p=0.03 respectively). Endothelin-1 overexpression was detected in 63% of the studied cases with a significant association with tumor size, FIGO stage and lymph node metastasis (p=0.009, p=0.002, p=0.02 respectively). High CD34 expression (MVD) was noted in 56% of the cases and associated with the tumor size, FIGO stage and lymph node metastasis (p<0.001, p<0.001, p=0.04 respectively). The three markers were significantly associated (p<0.05).Conclusion: EZH2, ET-1, and CD34 may act as biomarkers of aggressive cervical squamous cell carcinoma. They may contribute to the signaling pathway of angiogenesis. Therefore, they could potentially be used in targeted therapy.

Gastric cancer remains a big health problem in China. Gastric cancer cells contain a small subpopulation of cells that exhibit capabilities of differentiation and tumorigenicity. A putative explanation for ineffective therapy is the presence of cancer stem-like cells. Side population cells, which have cancer stem-like cells' property, are characterized by the high efflux ability of Hoechst 33342 dye. Side population cells have been isolated from gastric cancer cell lines in previous studies. The epithelial-mesenchymal transition is very important in the invasion and metastasis of epithelial-derived cancers. More and more studies showed that gastric cancer stem-like cells possess high invasive ability and epithelialmesenchymal transition property. A brief overview of the recent advancements in gastric cancer stem-like cells and epithelial-mesenchymal transition will be helpful for providing novel insight into gastric cancer treatment.
