Cancer drug therapy and stochastic modeling of “nano-motors”

Abstract: BackgroundControlled inhibition of kinesin motor proteins is highly desired in the field of oncology. Among other interventions, there exists “targeted chemotherapeutic regime/options” of selective Eg5 competitive and allosteric inhibitors, inducing cancer cell apoptosis and tumor regression with improved safety profiles.Research questionThough promising, such studies are still under clinical trials, for the discovery of efficient and least harmful Eg5 inhibitors. The aim of this research was to bridge the com… Show more

“…Kinesins play an important role in mitosis, and its abnormal expression or dysfunction can lead to malignant transformation of cells [14,[26][27][28]. Due to their important role in mitosis, kinesin family has become the target of chemotherapy intervention in malignant tumor [29].…”

“…These inhibitors arrest cells in mitosis, producing characteristic monoastral spindles [108,109,149], and lead to apoptosis in proliferative tissues either by binding to loop 5 or other allosteric sites and inhibiting ATPase activity [30,102,103,111,149,272]. To develop and test promising Eg5 inhibitors, different approaches are used, including in silico drug-design predictive tools that seek to bridge computational modeling with drug design and cancer therapy [259,273,274]. Based on these studies, several kinesin-5 inhibitors are currently in clinical trial [29,[275][276][277].…”

“…Kramer's seminal paper (1940) has been a prototype for a thermal activation process continually. Lately, activation in the presence of time varying fields has become a topic of vast significance because of the findings of many counterintuitive noise supported special effects, such as stochastic resonance (Gammaitoni et al 1998) or movement in Brownian motors (Sherin et al 2018).…”
