Cancer-associated rs6983267 SNP and its accompanying long noncoding RNA CCAT2 induce myeloid malignancies via unique SNP-specific RNA mutations

Abstract: The cancer-risk-associated rs6983267 single nucleotide polymorphism (SNP) and the accompanying long noncoding RNA in the highly amplified 8q24.21 region have been implicated in cancer predisposition, although causality has not been established. Here, using allele-specific transgenic mice, we demonstrate that overexpression leads to spontaneous myeloid malignancies. We further identified that is overexpressed in bone marrow and peripheral blood of myelodysplastic/myeloproliferative neoplasms (MDS/MPN) patients.… Show more

“…In recent years, lots of important trait-associated variants within lncRNAs have been identified using high-throughput methods such as genome-wide association studies (GWAS) and whole-genome sequencing [56][57][58] . Most of these variants are disease-associated variants in human and model animals such as mouse 59,60 .…”

“…Although the pyknons are located in about 225,000 places within the human genome, other more abundant genetic elements influence the structure of ncRNAs, and these are the SNPs. The examples of SNPs influencing the function of ncRNAs through the structural changes in ncRNAs are increasing at a rapid pace (Castellanos-Rubio et al 2016; Redis et al 2016;Bal et al 2017;Shah et al 2018).…”

“…In contrast to mRNAs, lncRNAs, have little or no translation potential [66] . LncRNAs can execute several functions working as scaffolds, guidance molecules, signaling pathways, and be decoys for other molecules [67][68][69] . Additionally, these long RNA molecules regulate gene expression by recruiting, to a specific DNA region, epigenetics modifiers or chromatin-remodelling complexes [66] .…”
