We have cloned and studied Hp-ECPN, an encephalopsin orthologue of the sea urchin Hemicentrotus pulcherrimus. Hp-ecpn cDNA was produced and found to contain a 1461-bp open reading frame that encodes 486 amino acids. Accumulation of Hp-ecpn mRNA and protein expression occurred at the 14 h postfertilization (hpf) swimming blastula stage and thereafter. The Hp-ECPN protein was N-glycosylated, and the amino acid sequence was similar to that of vertebrate encephalopsins. Whole-mount immunohistochemistry revealed the presence of Hp-ECPN in cells (ECPN cells) that appeared initially around the tip of the archenteron in 20 hpf early gastrulae. By the 54 hpf pluteus stage, ECPN cells had spread through the aboral ectoderm, and, by the eight-arm pluteus stage, were restricted to the tips of the larval arms and the posterior end of the body. The number of ECPN cells increased under conditions of continuous light, but decreased under continuous dark. Knockdown of Hp-ecpn mRNA using morpholino antisense oligonucleotides decreased the number of ECPN cells considerably, and inhibited the vertical swimming of the larvae. This suggested that Hp-ECPN plays a role in photosensitive larval swimming vertical migration. In adult tissues, the ECPN cells were detected exclusively in tube feet.

The present study is the ¢rst estimate for the sea urchin, Strongylocentrotus intermedius, of genetic parameters for growth and gonad traits during the entire reproductive period. 2165^5764 sea urchins were sampled from 74 to 120 families at six di¡erent growth stages. Six hundred individuals were sampled from 20 randomly chosen families at harvest. Coe⁄cients of variation for most growth and gonad traits were high (18.08^78.66%) suggesting a good variation foundation for breeding. Point estimate for heritabilities based on the sire components of variance were moderate to high for test height (0.24^0.39), test diameter (0.21^0.48) and body weight (0.16^0.49). Heritabilities of gonad wet weight, gonad index and gonad moisture at harvest were 0.17, 0.41 and 0.50 respectively (moderate or high). Gonad colour parameters, L Ã (lightness) and a Ã (redness) have a heritability of 0, while b Ã (yellowness) and colour di¡erence DE 1 and DE 2 have very similar heritabilities of 0.2, 0.18 and 0.21 respectively. Heritabilities of b-carotene concentration and content were 0.89 and 0.44 respectively. The genetic correlations between the two measured traits ranged from À 0.984 to 0.999. Gonad wet weight, L Ã , b Ã , bcarotene concentration and b-carotene content had signi¢cant positive genetic correlations with all the growth traits. However, gonad index, gonad moisture and colour di¡erences DE 1 and DE 2 had signi¢cant negative genetic correlations with growth traits. The present study indicates that growth, gonad yield and most gonad quality traits could be improved by selection with moderate to high potential. Indirect selection should be applied on gonad traits using growth traits as secondary traits. The present study provides important theoretical guidance for future selective breeding programmes for S. intermedius.

21 22We studied the reproductive cycle of the sea urchin Arbacia lixula in a subtidal population from 23Northeast Spain over four years using a gonadosomatic index and gonad histology. Our results 24show that the gonadosomatic index of A. lixula follows a seasonal cycle which peaks in May-July 25 and attains its lowest values in October-November every year. The time course of the 26 gonadosomatic index matched closely the photoperiod cycle. We also found a remarkable inter-27 annual variability in the maximum value of gonadosomatic index, which correlated with mean 28 water temperature during the gonad growth period (winter and spring). Gonad histology was also in 29 agreement with a single gametogenic cycle per year in this species. We explored the application of 30 circular statistics to present and analyse gonadal development data, which allowed us to adequately 31 handle the high intra-individual variability detected, with several developmental stages commonly 32 found within the same gonad. The picture that emerged is one of a gametogenic timing driven by 33 photoperiod, while the amount of reproductive output is determined by temperature. This is 34 coherent with the tropical origin of the species and lends support to recent warnings about an 35 increase in the abundance of this species in the Mediterranean as a result of global warming, with 36 associated increased impact potential in sublittoral communities. 37 38 Introduction 39
