Can MDM2 and CDK4 make the diagnosis of well differentiated/dedifferentiated liposarcoma? An immunohistochemical study on 129 soft tissue tumours

Abstract: Detection of MDM2/CDK4 protein overexpression by IHC can be used by pathologists to diagnose WDLPS and DDLPS. Considering a strong and diffuse immunostaining pattern in most of the neoplastic cells achieves the best results in identifying these tumours.

“…Consequently, detection of 12q amplified genes is widely used for distinguishing WDLPS/DDLPS from other tumors. This detection can be done by FISH [15], Q-PCR [42], or aCGH [4] and indirectly by immunohistochemistry [43,44]. MDM2 has early been identified as the main driver gene of the WDLPS/DDLPS amplicon since it is consistently amplified in all cases of WDLPS and DDLPS [10,43,45,46].…”

“…[8] Fluorescence in situ hybridization (FISH) for MDM2 amplification is a sensitive and specific tool which can be used for distinguishing difficult and doubtful cases of well-differentiated liposarcoma (WDLS). [1,9] In our case, the histopathological and immunohistochemical features were diagnostic and in favor of WDLS.…”

“…They are located in the extremities and retroperitoneum in about 70% of cases. [1,2,6,7,9] Paratesticular liposarcomas (PLS), however, are rare malignant tumors which account for approximately 3-7% of all paratesticular sarcomas. [1,3] Most of the available information on these tumors is derived from small series and case reports.…”

“…Immunostaining with murine double-minute type 2 and cyclin-dependent kinase 4 is useful to exclude liposarcoma. 4 The prognosis is excellent with no reported recurrences after complete resection. Because of the low prevalence of this unusual tumor, it is likely that few cardiologists will encounter this condition.…”
