Can Melissa officinalis improve chromatin structure and sperm parameters in a rat model of varicocele?

Abstract: Excessive production of reactive oxygen species is a central mechanism in the pathology of varicocele; Melissa officinalis (MO) is a medicinal plant from the Lamiaceae family that has antioxidant activity. In this study, we have investigated the potential therapeutic effect of MO on sperm parameters and chromatin structure in varicocelised rat. Male Wistar rats were randomly assigned to control, sham, varicocele, MO treatment and placebo groups. Rats in varicocele, MO treatment and placebo groups underwent lef… Show more

“…During the torsion period and resultant ischemia, ROS is assembled in the testicular tissue, while after surgery and torsion correction, reperfusion injury occurred due to reoxygenation of blood reopening (Gökçe et al, 2010). Low amount of ROS is necessary for normal spermatogenesis and fertility (Agarwal et al, 2009), however, when ROS creation exceeds the testicular defences, several reversible and irreversible problems will occur (Aktoz et al, 2010; Rezakhaniha et al, 2018). Therefore, free radicals are keystones in testicular I/R injury (Shi et al, 2016).…”

“…The analysis of spermatozoa was performed under a light microscope (Olympus BX‐50). Two hundred spermatozoa per animal were analyzed, which were classified according to the staining as follows: spermatozoa showing intact chromatin (light blue staining) and spermatozoa with uncompacted chromatin (dark blue or purple staining) 60 …”

“…Subsequently, the slides were washed three times in distilled water (1 min/each) and stained with 0.05% toluidine blue in 50% citrate buffer, pH 3.5 for 20 min. A total of two hundred spermatozoa per sample were evaluated under a light microscope at a 100× lens (Olympus BX‐51, Olympus Optical CO, Japan), and the chromatin quality was classified according to metachromatic staining of the head: spermatozoa showing light blue head (preserved chromatin) and spermatozoa showing dark blue or purple head (abnormal chromatin) 39 …”
