Calpain-1 Cleaves Rad21 To Promote Sister Chromatid Separation

Abstract: Defining the mechanisms of chromosomal cohesion and dissolution of the cohesin complex from chromatids is important for understanding the chromosomal missegregation seen in many tumor cells. Here we report the identification of a novel cohesin-resolving protease and describe its role in chromosomal segregation. Sister chromatids are held together by cohesin, a multiprotein ring-like complex comprised of Rad21, Smc1, Smc3, and SA2 (or SA1). Cohesin is known to be removed from vertebrate chromosomes by two disti… Show more

“…Leu-RAD21 is the Ctfragment of the SCC1/RAD21 subunit of the chromosome-associated cohesin complex. 147 Calpain-mediated generation of Leu-RAD21 contributes to the control of chromosome cohesion/segregation, together with processes that include the separase-mediated cleavage of the same RAD21 subunit 147,148,149,150 [see also Fig. 6(F)].…”

“…In addition to its roles in condensing chromatin during mitosis and meiosis, Condensin II has been implicated in chromatin condensation in a number of contexts during interphase (reviewed in Aragon et al, 2013; Hirano, 2012). While no direct link between [Ca 2+ ] i and Condensin II function has been established, the calcium protease Calpain-1 has been shown to cleave the α-kleisin subunit of the related Cohesion complex, promoting sister chromatid separation (Nasmyth and Haering, 2009; Panigrahi et al, 2011). Regulation of chromatin condensation via calcium by any of the above mechanisms is attractive because calcium is abundant in the cell and is rapidly mobilized in the time-frame in which we observed the initiation of chromatin decondensation following TCR stimulation.…”
